Cooperativity enables widespread role of low-affinity motifs in chromatin accessibility and increases regulatory potential.

Results show that low-affinity motifs can be accurately mapped, shape the properties of developmental enhancers, and likely play a widespread role in fine-tuning enhancers during evolution.
