Overcoming EGFR Resistance by Monovalent and Bident Inhibitors Targeting Cys775

This study establishes the therapeutic potential of an EGFR covalent inhibitor through unprecedented targeting of cysteine 775 and provides the first demonstration that dual cysteine engagement offers superior efficacy over conventional covalent inhibitors by delaying resistance.

PARP inhibition enhances the antitumor activity of HER3-DXd in non-small cell lung cancer

It is suggested that combining HER3-DXd with PARP inhibitors with a significant synergy is observed with PARP inhibitors, effective in both EGFR- and KRAS-mutated NSCLC models, effectively targeting diverse NSCLC subtypes.

Therapeutic targeting of RAS-mediated resistance in oncogene-driven lung cancer.

RAS alterations mediate resistance to targeted agents in approximately 10% of oncogene-driven lung cancer, and Rational combinations with novel RAS inhibitors are effective in preclinical models, providing the basis for their clinical investigation and extending the paradigm of precision oncology.
