Abstract

Enhancing innovation in herbicide discovery is imperative to combat weed resistance and safeguard global food security. To develop novel herbicide targeting dihydroorotate dehydrogenase (DHODH), a series of 3-carboxy-4-pyrazole amide lead scaffold bearing ester or acid substituents were designed and synthesized. Bioassay evaluation revealed that compound II-12 achieved an 81% growth inhibition activity against Setaria viridis at 135 g ai/ha in postemergence greenhouse assays, with herbicidal efficacy comparable or higher than that of several commercial products. Moreover, II-12 exhibited favorable crop safety toward four upland crops at the same dosage. In vitro enzyme inhibition assays demonstrated that II-12 inhibited SvDHODH with a half-maximal inhibition concentration value of 8.589 μM, while surface plasmon resonance analysis revealed an apparent dissociation constant Kd of 3.83 μM. Molecular docking combined with site-directed mutagenesis displayed that II-12 binds to SvDHODH through a critical H-bond interaction with Arg189. Collectively, these results identify II-12 as a promising DHODH-targeting herbicide candidate.

Discovery of Pyrazole-Benzimidazolin-2-One Derivatives as 4-Hydroxyphenylpyruvate Dioxygenase Inhibitors.

4-Hydroxyphenylpyruvate dioxygenase (HPPD) has become an attractive target for herbicide development owing to the slow development and low risk of resistance to its inhibitors. However, most available HPPD inhibitors exhibit limited crop selectivity and poor efficacy against grasses. Therefore, the exploration of novel...

Synthesis, Herbicidal Activity Evaluation, and Molecular Docking of Novel Acylthioureas as AHAS Inhibitors

Acetohydroxyacid synthase (AHAS, EC 2.2.1.6) is a core enzymatic target in agrochemical research for herbicide development and has been widely investigated in recent decades. To develop novel AHAS-targeted herbicides, twenty-three acylthiourea derivatives were synthesized via fragment recombination and bioisosteric rep...

Synthesis and Antifungal Evaluation of Isoquinoline‐3‐amide Derivatives Containing a Phenoxypyridine Moiety for Plant Protection

To discover novel green antifungal candidates, 20 3‐acylaminoisoquinoline derivatives were designed and synthesized by introducing the active fragment phenoxypyridine. Their structures were verified by 1H NMR, 13C NMR, and HRMS. The structure of compound X9 was further confirmed via X‐ray crystallography. Their antifun...

Rational Design of Novel Tetrazolinone-Pyrazine Derivatives: Synthesis, Antifungal Activity, and Mechanism of Action

G143A mutation-driven target-site resistance severely impairs the field efficacy of traditional Quinone outside inhibitors (QoIs). To solve this agricultural crisis, novel tetrazolinone-pyrazine derivatives were rationally designed and synthesized by integrating potentially resistance-breaking tetrazolinone toxophore...
