PA1/JAM3 dual-gene methylation testing demonstrates high diagnostic accuracy for detecting high-grade cervical intraepithelial neoplasia and cervical cancer, supporting its potential role as a triage biomarker in high-risk human papillomavirus positive women.
Abstract
Background: DNA methylation is an emerging biomarker for cervical cancer screening. This study aimed to evaluate the diagnostic performance of paired box gene 1/junctional adhesion molecule 3 (JAM3) dual-gene methylation analysis for detecting high-grade cervical intraepithelial neoplasia and cervical cancer, and to explore its potential utility as a triage biomarker for high-risk human papillomavirus positive women. Methods: A systematic search was conducted across 5 databases, including PubMed, for studies on PAX1/JAM3 methylation testing in cervical cancer screening. Following quality assessment via the QUADAS-2 tool, statistical analyses were performed using Meta-Disc 1.4 and Stata 17 software. Results: Eight studies involving 7494 participants were included in the meta-analysis. For diagnosing cervical intraepithelial neoplasia grade 2 and above the pooled sensitivity was 0.81 (95% CI: 0.73–0.88), specificity was 0.95 (95% CI: 0.94–0.96), and the area under the curve was 0.96. For diagnosing cervical intraepithelial neoplasia grade 3 and above the pooled sensitivity was 0.85 (95% CI: 0.75–0.92), specificity was 0.88 (95% CI: 0.86–0.89), and the area under the curve was 0.90. Conclusions: PAX1/JAM3 dual-gene methylation testing demonstrates high diagnostic accuracy for detecting high-grade cervical intraepithelial neoplasia and cervical cancer, supporting its potential role as a triage biomarker in high-risk human papillomavirus positive women.
OBJECTIVE
To perform a systematic review and meta-analysis on the utility of circulating tumor DNA (ctDNA) in serum as a biomarker for post-treatment surveillance of human papillomavirus (HPV)-positive oropharyngeal squamous cell carcinoma (OPSCC), given the improved treatment response and 10% to 20% recurrence rate in...

Methylation biomarkers of six tumor suppressor genes (astrotactin 1, distal-less homeobox 1, integrin subunit α 4, relaxin family peptide receptor 3, SOX17 and zinc finger protein 671) were evaluated for their diagnostic efficacy in identifying cervical precancerous lesions and cervical cancer (CC). A total of 283 case...

High-risk human papillomavirus (Hr-HPV) testing is highly sensitive for cervical cancer screening but lacks specificity, leading to unnecessary colposcopy referrals. Host-gene methylation and HPV viral load capture different aspects of disease risk and may support triage. In this prospective real-world triage cohort, 1...
The diagnostic performance of PAX1/JAM3 methylation for high-grade cervical lesions differs distinctly between premenopausal and postmenopausal women undergoing colposcopy, and should be considered when applying methylation biomarkers, particularly as a valuable auxiliary tool to improve diagnostic accuracy in elderly...

Background/Objectives: Excisional treatment is effective for cervical intraepithelial neoplasia grade 2 or 3 (CIN2–3), but residual or recurrent high-grade disease remains clinically important. This focused systematic review evaluated post-treatment high-risk human papillomavirus (hrHPV) testing as a test of cure and c...

Lung cancer remains the leading cause of cancer-related mortality worldwide, with poor prognosis in advanced stage diseases. Although early diagnosis has the potential to improve patient outcomes, current diagnostic methods remain suboptimal, highlighting the need for accurate molecular biomarkers. We systematically re...
