Abstract

Background

Allostatic load is a multisystem composite measure that quantifies the cumulative physiological wear and tear caused by chronic stress, often traceable to early life, where adverse childhood experiences (ACEs) serve as a key risk factor.

Objective

This study aims to identify distinct co-occurring patterns of ACEs, examine their relationship with allostatic load later in life, and investigate potential buffering mechanisms.

Methods

Data were from 2090 adults (age range 25-84, M = 53.019, SD = 12.556; 45.3% male) in the Midlife in the United States (MIDUS) database. ACEs were assessed using the validated Childhood Trauma Questionnaire; allostatic load was operationalized as a composite index with 24 biomarkers across cardiovascular, immune, neuroendocrine, and metabolic systems.

Results

Five distinct profiles emerged: no adversity (50.96%), mild emotional neglect (7.27%), mild emotional neglect and sexual abuse (7.42%), emotional neglect and emotional abuse (25.26%), and multiple adversity (9.09%). Compared with no adversity, emotional neglect and emotional abuse (b = 0.138, SE = 0.063, p < 0.05) and multiple adversity (b = 0.345, SE = 0.090, p < 0.001) were associated with higher allostatic load later in life. Purpose in life was associated with lower allostatic load (b = -0.009, SE = 0.004, p < 0.01) but did not serve as a buffer.

Conclusions

ACEs frequently co-occur, and individuals exposed to multiple types of adversity are at the greatest risk of physiological dysregulation. Purpose in life may function more as a promoting factor than as a protective buffer in associations between ACEs and physiological functioning.

733. Effects of early life adversity on stress markers and emotion circuitry in adults with a history of suicide attempts

Abstract Background Early life adversity (ELA) has been highlighted as strong distal factor for emergence of suicide behaviors (SB), however not all persons that experience ELA will engage in SB. Dispositional factors, such as stress system responsivity, emotion dysregulation, and brain structure and connectivity are h...

The Unique and Joint Associations of Childhood and Recent Life Stressors on Chronic Pain Symptoms

Background/Objectives: Early life stress is a well-established predictor of chronic pain; however, less is known about whether early and recent life stressors interact to influence chronic pain. The purpose of the present study was to examine the unique and joint associations of early and recent life stressors on chron...

Proteomic insights into childhood adversity: Exploring biological mechanisms and cognitive outcomes in the CARDIA study

Adverse childhood experiences (ACEs), including abuse, neglect, and household dysfunction, are among the most consequential adverse social experiences of early life and carry elevated risk of chronic disease and cognitive impairment in adulthood through biological pathways that are poorly understood. We examined the me...

Psychological Resilience and End-of-Life Symptom Burden: Evidence From a National Cohort Study.

BACKGROUND Distressing symptoms are common in the last year of life and contribute substantially to suffering for older adults and their families. While psychological resilience has been linked to better aging outcomes, its association with end-of-life symptom burden is not well characterized. METHODS We conducted a...

Stress sensitization: Does childhood adversity modify trauma effects on memory?

Background Trauma across the lifecourse is implicated in adverse health outcomes, yet whether earlier adversity shapes vulnerability to effects of later trauma on cognitive decline, an important contributor to Alzheimer's disease, remains unclear. Objective We examined the impact of adult incident traumatic experiences...

Refining Allostatic Load Measurement: Mortality Associations of an Empirically Derived Three-Component Score in the Women's Health Initiative.

Allostatic load (AL) is a multisystem index of physiological dysregulation due to chronic stress. Although widely applied, its operationalization remains inconsistent, limiting reproducibility and equity. Streamlined indices based on common biomarkers may enhance feasibility across settings. Using data from 150 293 pos...
