Abstract

Simple Summary Aging is the key risk factor for a variety of diseases, such as heart disease, diabetes, and dementia. One of the hallmarks of aging is cellular senescence, a condition where cells stop dividing but remain metabolically active, thus becoming detrimental to the surrounding tissues. This review focuses on a protein known as SIRT1, a molecular guardian of our cells. SIRT1 plays a critical role in maintaining mitochondria health and in suppressing inflammation. Levels of SIRT1 decrease with age, which leads to mitochondrial dysfunction and the release of mitochondrial DNA into the cytoplasm, triggering inflammation. This inflammation further reduces SIRT1 levels, creating a cycle that accelerates cellular aging. This review discusses the most recent and relevant SIRT1, mitochondrial and inflammatory-related findings and highlights prominent strategies, such as increasing SIRT1 activity, to promote healthier and longer lives.

Pro-Longevity Role of SIRT6 to Promote Life Span and Delay Ageing – An Integrated Review Towards Healthy Ageing

Ageing is an outcome of insufficient maintenance of the internal environment. Over time, each component of the internal environment changes from the molecular level to the organismal level to maintain homeostasis. These changes occur through several biological pathways, such as adenosine monophosphate-activated prote...

The Role of Cellular Senescence in Chronic Lung Diseases: Emerging Mechanisms and Translational Perspectives: A Narrative Review

Cellular senescence is one of the major risk factors for the onset and progression of chronic pulmonary diseases. Cellular senescence can be induced by diverse stressors, including genotoxic damage, oncogenic signaling, and therapeutic interventions. These senescent cells communicate via the release of multiple inflamm...

SIRT1 in Neurodegenerative Diseases: Molecular Mechanisms, Disease Relevance, and Therapeutic Potential

Neurodegenerative diseases are characterized by progressive protein aggregation, mitochondrial dysfunction, neuroinflammation, and cognitive decline, yet effective mechanism‐based interventions remain limited. Sirtuin 1 (SIRT1), an NAD+‐dependent deacetylase, has emerged as a multifunctional regulator linking stress ad...

Therapeutic potential of MitoQ, a mitochondria-targeted antioxidant, in age-related physiological dysfunction.

Aging is an inevitable and progressive physiological process marked by a decline in cellular function, accumulation of somatic mutations, decreased ability to maintain homeostasis, and increased tissue and organ dysfunction. These changes contribute to the onset and progression of various age-related diseases. Accumula...

A systematic evaluation of SIRT6 as a transcriptomic biomarker of aging

SIRT6 is a NAD+-dependent sirtuin that plays central roles in chromatin regulation, DNA repair, telomere maintenance, metabolic homeostasis, and inflammatory control. Although SIRT6 have long been implicated in aging because of its association with several hallmarks of aging, the available evidence remains largely cont...
