TL;DR

Evidence is provided that the Drosophila orthologue of FOXA, known as fkh, is instrumental in preventing programmed cell death in Drosophila Malpighian tubules (MTs) during metamorphosis and plays an essential role in the survival of MTs.

Abstract

Forkhead (fkh) transcription factor derives its name from the distinctive appearance of the DNA-binding motifs when attached to DNA. In mammals, they are grouped into seven families from FOXA to FOXS. FoxA subfamily participates in several essential functions, including the development of various organs, the regulation of senescence, proliferation, and metabolic homeostasis. In this study, we provide evidence that the Drosophila orthologue of FOXA, known as fkh, is instrumental in preventing programmed cell death in Drosophila Malpighian tubules (MTs) during metamorphosis. This characteristic is unique to Malpighian tubules when compared to several other larval tissues. We also demonstrate its role in the immune function of the MTs, as its depletion in MTs suppresses the expression of Anti-Microbial Peptides (AMPs). Furthermore, reduced expression of NF-kB transcription factor, Relish and Dorsal, could contribute to the reduced synthesis of AMPs. Fkh also regulates the fluid secretion of the MTs and its absence disrupts the ion transporters thus affecting fluid homeostasis. Our results demonstrate that fkh plays an essential role in the survival of MTs, without that the MTs cannot survive and fail to mount an effective immune response.

Nervous System Integrity in Drosophila: Essential and Dosage-Sensitive Roles of Hsp110.

The Hsp70 chaperone system is central in maintaining cellular proteostasis. In eukaryotes, its ATP/ADP nucleotide exchange cycle is stimulated by co-chaperone Hsp110, a divergent Hsp70 superfamily member and a modifier of protein misfolding disorders. Hsp110 chaperone functions and pathway interactions have been extens...

Actin depolymerization factor (ADF) moonlighting: Nuclear immune regulation by interacting with WRKY transcription factors and shaping the transcriptome.

Remodeling of the actin cytoskeleton is a critical process for plant immunity, essential for the transport, activation, and stabilization of immune-regulatory molecules and organelles. In this process, actin depolymerization factors (ADFs) function as key players through severing and depolymerizing actin microfilaments...

Drosophila heat shock factor (HSF) regulates developmental growth by maintaining the basal expression of HSP83/HSP90.

The heat shock transcription factor HSF1 is best known as a master regulator of the proteotoxic stress response, yet its functions remain incompletely understood. In Drosophila melanogaster, heat shock factor (HSF) is essential for viability, but the mechanisms by which it promotes development are unclear. Here, we sho...

An atlas of the C. elegans starvation response reveals that transcription is necessary to initiate transcriptional silencing in quiescent primordial germ cells

It is revealed that transcription of aak-1/AMPK is necessary to silence transcription in quiescent PGCs, and it is validated PGC transcription of aak-1/AMPK, and it is showed that zygotic aak-1/AMPK is required for PGC chromatin hyper-compaction, supporting reproductive success upon recovery.

FBXL21 regulates diurnal proteostasis in skeletal muscle by targeting DNAJB6 and client proteins.

Circadian regulation of proteostasis, a key determinant of muscle health, remains poorly understood. Here, we identify DNAJB6, an Hsp40 (DnaJ) co-chaperone, as a substrate of the circadian E3 ligase FBXL21. FBXL21 mediates the ubiquitination-dependent proteasomal degradation of both DNAJB6 and its client proteins, incl...
