TL;DR

A potential role for TH9 cells and their signature cytokine IL-9 in the antiviral immune response in KTRs is suggested, although further research is needed to fully elucidate their protective mechanisms against viral infections.

Abstract

Human cytomegalovirus (HCMV) is a frequent complication in kidney transplant recipients (KTRs), often impacting immune regulation. TH9 cells, a subset of CD4+ T cells, are characterized by their secretion of interleukin-9 (IL-9). This study aimed to analyze the phenotypic profile of TH9 cells and their associated cytokine expression in KTRs, and to evaluate mRNA levels of IL-4 and transforming growth factor-β (TGF-β), key cytokines involved in TH9 differentiation. Ten HCMV+ and 10 HCMV- KTRs, along with 10 age- and sex-matched healthy controls, were enrolled. HCMV viral load was quantified using TaqMan real-time polymerase chain reaction. Flow cytometry was used to assess surface markers (CCR6+CCR4-IL-4Rα+CD4+) and intracellular IL-9 expression in TH9 cells. Gene expression levels of IL-4 and TGF-β were also assessed. Surface staining revealed a significantly higher frequency of CD4+CCR4- and CD4+CCR6+ T cells in HCMV- KTRs compared to HCMV+ patients. Conversely, the overall frequency of CD4+ TH9 cells was elevated in HCMV+ KTRs. Intracellular staining demonstrated a significant increase in CD4+IL-9+ and CD4+IL-4Rα+ T cells in the HCMV+ group. Additionally, mRNA expression levels of IL-4 and TGF-β were markedly higher in HCMV+ KTRs than in HCMV- counterparts. These findings suggest a potential role for TH9 cells and their signature cytokine IL-9 in the antiviral immune response in KTRs. While TH9 cells may contribute to HCMV-related immune modulation, further research is needed to fully elucidate their protective mechanisms against viral infections.   Keywords: ; ; ;

Abatacept Reduces CD319+ (SLAMF7) Cytotoxic T Cells and Cytokine Production in Systemic Sclerosis.

Abatacept selectively modulates CD319/SLAMF7-expressing cytotoxic T cells and suppresses associated pro-fibrotic and pro-inflammatory cytokines, particularly in the fibroproliferative subtype, where CD319+ T cell reductions correlate with clinical improvement.

Eltrombopag-associated remodeling of the MAIT, MR1+ and γδ T cells in pediatric immune thrombocytopenia.

Pediatric immune thrombocytopenia (ITP) is an autoimmune cytopenia characterized by immune-mediated platelet destruction and impaired thrombopoiesis. Mucosa-associated invariant T (MAIT) cells are innate-like T cells activated by MR1-presented microbial metabolites and inflammatory cytokines, but their role in pediatri...

Intragraft FOXP3+ Cells and Continuous Banff Indices in T Cell-Mediated Kidney Allograft Rejection

Background and Objectives: FOXP3+ (Forkhead box P3) regulatory T cells contribute to immune tolerance after kidney transplantation, but their role in T cell-mediated rejection (TCMR) remains controversial. This study investigated the association between intragraft FOXP3+ cells and the recently introduced Banff-derived...

IL-27 induces a cytotoxic state in CD4+ T cells distinct from the Th1 lineage

CD4+ cytotoxic T lymphocytes (CD4-CTLs) are understudied immune mediators with ambiguous origins. Despite expressing RUNX3, granzyme B (GZMB), and perforin (PRF1), CD4-CTLs are frequently classified as Th1 extensions due to shared interferon-γ (IFNγ) and T-BET expression. Here, we identify interleukin-27 (IL-27) as a i...

CD38+ T-Cell Count Is an Immunologic Phenotype Associated With Blood Pressure.

BACKGROUND T lymphocytes play a crucial role in the development of hypertension and associated end-organ damage. CD38 is a well-established surface marker for T-cell activation. However, clinical evidence linking CD38+ T cells, or other specific T-cell subsets, with blood pressure (BP) changes remains limited. We there...

The role of CD4+ T cell differentiation in cytomegalovirus-associated parahippocampal volume reduction and depressive symptoms: a dual-sample study.

Cytomegalovirus (CMV) establishes lifelong infection and promotes an immunosenescent T cell profile characterized by depletion of naïve cells and expansion of terminally differentiated effector memory cells re-expressing CD45RA (TEMRA). We examined whether CMV-related T cell differentiation was associated with brain st...
