Abstract

Nitrate stands as a primary nitrogen (N) source for plants across diverse natural habitats and agricultural systems. It serves as both a vital nutrient and a key signaling molecule crucial for plant growth and development. The NIN-like protein 7 (NLP7) functions as a central transcription factor and intracellular nitrate sensor in Arabidopsis thaliana (Arabidopsis), yet its orthologs remain uncharacterized in Chinese cabbage (Brassica rapa subsp. pekinensis), a globally important vegetable crop. In this study, the BrNLP7.1 gene was identified based on its homology with NLP7 in Arabidopsis, and its role in mediating nitrate assimilation and signal transduction was characterized. Ectopic overexpression of BrNLP7.1 in the Arabidopsis nlp7 mutant rescued the stunted growth phenotype, restored nitrate content and nitrate reductase activity, and recovered the expression of nitrate-responsive gene. Yeast one hybrid assays and transient expression assays demonstrated that BrNLP7.1 directly binds to the nitrate-responsive cis-element in target gene promoters and activates transcription. Furthermore, microscale thermophoresis analysis revealed that the N-terminal region of BrNLP7.1 directly binds nitrate. Collectively, these findings establish BrNLP7.1 as a functional ortholog of NLP7 involved in nitrate-responsive signaling.

Functional Characterization of the Oat (Avena sativa L.) TCP Transcription Factor AsTCP38 Reveals Its Role in Low-Nitrogen Stress Tolerance

It is suggested that AsTCP38 may participate in low-nitrogen responses and provide a basis for further functional studies in oat and direct regulatory targets and the contribution of AsTCP38 to low-nitrogen adaptation in oat remain to be established.

CIPK11 Regulates Plant Adaptation to Nitrate Availability in Arabidopsis

Nitrate (NO3−) is an essential nitrogen source and an important signaling molecule that regulates plant growth and development in response to nitrogen availability. Although several CBL-interacting protein kinases (CIPKs) have been implicated in nitrate signaling, the role of CIPK11 (PKS5) in nitrate availability respo...

NRT1.1 as central signaling hub orchestrating plant growth and stress trade-offs

Plants must continuously balance nitrogen-dependent growth with adaptation to environmental stresses. The nitrate transporter AtNRT1.1 in Arabidopsis thaliana and its rice ortholog OsNRT1.1B have been reported to integrate nitrogen- and stress-related signals in their respective species. This review synthesizes how NRT...

Genome-Wide Analysis of the PYL Gene Family and Its Expression Dynamics in Response to Abscisic Acid in Tomato

The plant hormone abscisic acid (ABA) plays a crucial role throughout the plant life cycle and in adaptive responses to environmental stresses. The pyrabactin resistance 1-like (PYR/PYL/RCAR) proteins act as key regulators in the ABA signal transduction pathway by functioning as direct receptors for ABA. Although PYL g...
