Abstract
We report Bessel-beam-based Fourier light-field microscopy (Bessel-FLFM), an extended-depth-of-field (DOF) snapshot volumetric imaging method. By integrating annular pupil modulation into Fourier light-field detection, Bessel-FLFM employs an annular microlens array to generate Bessel-like point spread functions, enabling substantial DOF extension for high-resolution volumetric imaging. We further introduce radiality-analysis-based Richardson–Lucy deconvolution for high-resolution three-dimensional reconstruction. Experiments with fluorescent beads demonstrate a 16 μm DOF, with lateral and axial resolutions of approximately 350 and 500 nm, respectively. High-speed volumetric tracking of freely diffusing beads achieves an imaging rate of 100 Hz. Multicolor imaging of biological samples further validates the applicability of Bessel-FLFM for snapshot volumetric fluorescence imaging at subcellular resolution.
Summary Achieving isotropic high-resolution three-dimensional imaging across a large field of view (FOV) remains a key challenge in two-photon light-sheet microscopy (TPLSM). To address this limitation, we developed a TPLSM system that stitches multiple short Bessel beams into a large, ultrathin illumination light shee...

Two-photon microscopy (2PM) constitutes a key-enabling technology for imaging in weakly scattering specimens, delivering high-contrast signals through the quadratic dependence of fluorescence on excitation intensity. Extending 2PM to widefield or scan-less formats is attractive for rapid functional imaging. To maintain...

An electrically modulated dual-focus metalens-based photoacoustic microscopy (meta-PAM) system designed to enhance axial coverage while preserving high lateral resolution in three-dimensional imaging is presented, demonstrating its effectiveness by imaging a chemically induced corneal burn in rat eyes.

Light-field microscopy enables snapshot volumetric imaging, but its information rate is constrained by both optical encoding and detector readout architecture. Here we develop a task-dependent Fisher-information framework that evaluates optical encoders relative to the detector resource limiting acquisition throughput....
Fourier light-field microscopy (FLFM) enables single-shot 3D imaging by placing a microlens array at the Fourier plane conjugate to the objective's aperture stop, yielding a spatially invariant 3D point-spread function and artifact-free reconstruction. This performance depends critically on a near-diffraction-limited w...
