The development and validation of a sensitive and selective LC–MS/MS method for the estimation of venlafaxine impurity‐H in desvenlafaxine succinate API is described and is suitable for routine quality control and regulatory compliance applications.
Abstract
Desvenlafaxine succinate, the major active metabolite of venlafaxine, is widely prescribed for the treatment of major depressive disorder. During synthesis and process optimization, structurally related impurities such as venlafaxine impurity‐H may be formed. Due to its structural similarity and potential toxicological concern, accurate trace‐level quantification of this impurity in desvenlafaxine succinate active pharmaceutical ingredient (API) is essential. The present study describes the development and validation of a sensitive and selective LC–MS/MS method for the estimation of venlafaxine impurity‐H in desvenlafaxine succinate API. Chromatographic separation was achieved on a C18 column under gradient elution using aqueous formic acid and methanol. Detection was performed in positive electrospray ionization mode using multiple reaction monitoring (MRM). The method was validated in accordance with ICH Q2(R2) guidelines and demonstrated excellent linearity (
r
≥ 0.995), acceptable precision, satisfactory recovery across the validated range, and high sensitivity. The developed method is suitable for routine quality control and regulatory compliance applications.
Nitrosamine drug substance–related impurities have been a central focus of regulatory scrutiny due to their potential mutagenic and carcinogenic effects. Among these,
N
‐nitroso tizanidine (
N
‐NTZD) is a possible impurity that may form during the manufacturing, processing, or storage of tizanidine (TZD) drug s...
A rapid and highly sensitive LC-MS/MS method was developed and validated for the determination of the nitrosamine drug substance-related impurity N-nitroso-desmethyl sumatriptan (NNDS) in sumatriptan succinate injection. Separation was achieved on an Inertsil ODS-3 column using gradient elution with 0.1% formic acid in...
In the process of making vildagliptin (VG) API, one of the nitrosamine groups found in the drug is
N
‐nitroso vildagliptin amide (NVA). This led to the development of a multi‐reaction monitoring (MRM) approach that is both sensitive and reliable for quantitatively assessing NVA impurity in VG API. Using isocratic...
Application of the validated method to commercial herbal weight-loss supplements successfully identified products adulterated with sibutramine and desmethylsibutramine, and all positive findings were independently confirmed by high-resolution mass spectrometry.
