The optimized Aceclofenac gel avoids the first pass metabolism, and it showed a significantly enhanced permeability through the porcine skin and good stability.

Abstract

Aceclofenac is a non-steroidal anti-inflammatory drug used to treat pain and inflammation caused by musculoskeletal disorders. Oral administration of Aceclofenac is associated with various gastrointestinal side effects like dyspepsia, abdominal pain, or discomfort. To reduce systemic side effects and to improve patient compliance, a topical gel was formulated using the Quality by Design approach. The Aceclofenac gel was prepared by using Carbopol 974P as a polymer and optimized by custom design. The design suggested 8 runs for 3 factors (Carbopol 974P, limonene and PEG 400) and 2 responses (viscosity, in vitro drug release). The 8 different formulations were prepared and evaluated. The optimized topical gel was characterized for homogeneity, pH, spreadability, viscosity, drug content, in vitro drug release, ex vivo permeation, and stability studies. The optimized Aceclofenac gel formulation predicted global desirability was 0.9980; in-vitro and ex vivo studies were found to be 84.64% and 77.42%. The drug release kinetics were analyzed using zero-order drug kinetics, the highest correlation of 0.999. A stability study was performed, and the shelf life was predicted to be 23.8 months using JMP statistical software. The optimized Aceclofenac gel avoids the first pass metabolism, and it showed a significantly enhanced permeability through the porcine skin and good stability.

Development and evaluation of microemulsion foam formulation for topical delivery of betamethasone dipropionate.

Betamethasone dipropionate (BDP) is widely recognized as a therapeutic agent for psoriasis. However, the current topical formulations exhibit certain limitations, including significant systemic absorption and substantial residue on the skin surface post-application. Thus, this study aimed to develop a novel microemulsi...

Preparation and Evaluation in vitro of Usnic Acid Emulgel with Varying Concentrations of Propylene Glycol

Usnic acid is one of the secondary metabolites found in lichens. Usnic acid has anti-inflammatory activity and is classified as class 2 in the Biopharmaceutical Classification System (BCS), with low solubility but high permeability. Therefore, a suitable delivery system is needed to enhance the release and penetration...

Design of Eudragit E100 Films Incorporating Lipoic Acid as a Permeability-Enhancing Platform for Prednisolone Delivery in the Treatment of Ophthalmic Diseases

Prednisolone (PRED), a widely used corticosteroid for the treatment of ocular inflammation, has limited bioavailability due to its poor water solubility. In this study, Eudragit E100-based films incorporating polyethylene glycol 400 (PEG) and α-lipoic acid (ALA) as plasticizers were developed and characterized to impro...

DoE-oriented optimization of a chitosan-based nanoemulgel of clobetasol propionate for therapeutic intervention in psoriasis

Psoriasis is an inflammatory skin disease caused by immune-mediated inflammation. Clobetasol propionate (Cp), a steroid, reduces inflammation by binding the glucocorticoid receptor. This study aimed to optimize a Cp-loaded nanoemulsion (Cp-NE) with a focus on its cytocompatibility with HaCaT keratinocytes and physicoch...
