Abstract

Post-transcriptional gene silencing (PTGS) is a conserved RNA-based defense mechanism that protects plants against viruses and other invasive nucleic acids (Tan et al. 2020). During PTGS, double-stranded RNAs (dsRNAs) are processed by DICER LIKE 4 (DCL4) and DCL2 into 21-and 22-nucleotide small interfering RNAs (siRNAs), respectively, which are incorporated into ARGONAUTE (AGO)-containing RNA-induced silencing complexes (RISCs) that recognize and degrade homologous transcripts. In addition to local silencing, PTGS also generates mobile silencing signals that spread systemically throughout the plant (De Meo et al. 2025)

Circular RNAs as central regulators of RNA stability.

Circular RNAs (circRNAs) are a class of covalently closed, single-stranded RNAs generated predominantly through back-splicing of pre-mRNAs. Unlike their linear counterparts, circRNAs lack free 5' and 3' termini, rendering them intrinsically resistant to exoribonucleolytic degradation and conferring exceptional stabilit...

Flipons, Noncoding Transcripts and Retroelements: A Universal Host Defense

Encoding genetic information involves both structure and sequence. Non-protein-coding (NCR) RNAs modulate host immune responses by guiding and assembling stress-specific protein effectors, with flipons enabling dynamic transitions between states. Small NCRs transcribed by both RNA Polymerases 2 and 3 normally modulate...

Cellular signal-responsive m5C editing by an inducible split CRISPR platform.

RNA 5-methylcytidine (m⁵C) modification plays an essential role in regulating RNA metabolism and functions in cellular processes. Tools achieve temporal and transcript-specific m⁵C editing for functional studies are still limited. Furthermore, methods enabling m⁵C editing, triggered by specific cellular signals, can co...

Argonaute 15 regulates immunity of potato to filamentous pathogens through transposon-derived small RNAs

Canonical ribonucleases such as Argonaute (AGO) participate in RNA-based gene regulatory processes. They are important for small RNA (sRNA) association and the formation of RNA-induced RNA silencing complexes. Potato (Solanum tuberosum) has 14 AGO-encoding genes, including the Solanum-specific StAGO15, which is activat...

Noncanonical Functions of DICER-Mediated Pathways.

This review delineates the classical, RNAse III-dependent small RNA processing activities from an expanding set of noncanonical functions in which Dicer operates independently of RNAi outputs, and proposes an integrated conceptual framework in which Dicer acts as a multifunctional regulator of chromatin architecture an...

Multiple Argonaute complexes orchestrate epigenetic silencing in the Arabidopsis germline

Small RNAs (sRNAs) are widespread across diverse flowering plant species and play important roles regulating reproduction and environmental responses. However, how sRNAs exert their function is largely unknown, likely due to their typical origin in non-genic regions, the lack of distinct gene targets, and the sparse ch...
