TL;DR

The EC test presents a promising approach for rapidly assessing spore viability and longevity, and lower initial EC (EC0) values predicted greater initial spore viability and longer longevity, likely reflecting differences in spore maturity.

Abstract

Abstract Premise Ex situ spore banks are essential for fern conservation and research, yet traditional germination tests for assessing spore quality are time consuming and labor intensive. We explored whether electrical conductivity (EC), a rapid proxy for seed vigor, could offer an efficient alternative for evaluating fern spore quality. Methods Using chlorophyllous spores of Plenasium banksiifolium, we developed a microscale EC protocol (0.01‐g spores, 1‐h immersion) to track EC and germination over eight weeks of storage. Spore longevity (p 50) was estimated via probit analysis, and correlations among EC, viability, and longevity were assessed. Results Fresh spores from different individual plants showed varied initial EC (EC0) values, with lower EC0 values associated with longer longevity (p 50). During storage, EC values increased over time, while germination percentage declined. The relative EC ratio (ECi/EC0) showed significant negative correlations with germination percentage, with a ratio of ~2 indicating 50% viability loss and ~3 signaling complete viability loss. Discussion The EC test presents a promising approach for rapidly assessing spore viability and longevity. Lower initial EC (EC0) values predicted greater initial spore viability and longer longevity, likely reflecting differences in spore maturity. The relative ECi/EC0 ratio effectively tracked viability loss, supporting its use in improving spore bank management.

Morphometric Characterization and Germination Potential of Frangula sphaerosperma Seeds

Abstract This study explores the propagation of Frangula sphaerosperma, a species relevant for ecological restoration, by characterizing its seed and biometric traits (length, width, and shape indices) and evaluating germination at 20, 25, 30 and 35 °C. Germination percentage, germination speed index, average germinati...

Electrical Conductivity Estimates of Normal Germination Percentages of Seed Lots Belong to The Same and Different Cultivars in Cabbage

Electrical conductivity testing is a widely used, rapid, and non-destructive method to assess seed vigor and predict germination potential. This study investigated the potential of electrical conductivity to predict normal germination percentages in cabbage (Brassica oleracea L.) seed lots of varying genetic background...

Infrared and CO2-based assessment of seed viability in Lafoensia pacari A.St.-Hil. (Lythraceae)

Rapid methods for assessing seed viability were evaluated using Lafoensia pacari as a biological model, focusing on infrared emission (IRe) and carbon dioxide (CO₂) as potential indicators. Seed Morphology, dimensional traits, thousand-seed mass, water content, temperature profiles (air, seeds, and substrates) via IRe,...

Influence of Contained Cold Plasma Processing on Multi‐Species Sward Seeds Germination and Plant Growth Performance Properties

Multi‐species Swards (MSS) grasslands are recognized for mitigating agriculture‐associated emissions, but the establishment and maintenance of MSS profiles remain challenging due to variable germination and early seedling growth. In this study, cold plasma priming of MSS was evaluated by comparing germination and s...
