TL;DR

Results indicate that snRNA-seq recapitulates the dynamic changes associated with injury as previously reported using single-cell methods and outperforms these approaches in the representation of cell types that are sensitive to processing, thereby highlighting its utility for high resolution analysis of heterogenous tissues such as lung.

Single-cell and single-nucleus transcriptomics in ischemic stroke: cellular mechanisms and therapeutic implications

This review systematically integrates current evidence from sc/snRNA-seq studies, with a primary focus on brain-intrinsic mechanisms after ischemic stroke, to provide a structured synthesis of how sc/snRNA-seq has advanced the understanding of ischemic stroke biology.

Integrated single-cell and spatial transcriptomic analyses identify a unique subset of pro-fibrotic macrophage in kidney fibrosis.

INTRODUCTION Acute kidney injury (AKI) is a severe clinical syndrome associated with high morbidity and mortality, and it possesses high risk of progression to chronic kidney disease (CKD). Renal infiltration of inflammatory cells, particularly macrophages, plays a key role in driving the progression from AKI to CKD....

Dynamic orchestration of structural and immune cells in acute Pseudomonas aeruginosa pneumonia revealed by time-resolved single-cell and spatial transcriptomics

Background Pseudomonas aeruginosa pneumonia triggers a complex, dynamic host response, yet the cellular coordination of inflammation and repair remains poorly defined. Methods We performed time-resolved single-cell RNA sequencing (scRNA-seq) on murine lungs across five timepoints (0–96 h post-infection) and integrated...

Single Cell Cytometry of Mouse Brain Tissue

Suspension mass cytometry can measure 50 or more markers per cell and is not impacted by cellular autofluorescence, making it an appealing platform for comprehensive phenotyping of single cells in tissue with high innate fluorescence, such as brain. However, this organ is challenging to reduce to a single cell suspensi...

A NET-competent neutrophil subpopulation drives early brain injury after subarachnoid hemorrhage

Introduction Early brain injury (EBI) following subarachnoid hemorrhage (SAH) critically determines neurological outcomes, yet the cellular drivers remain unclear. Methods To systematically dissect acute pathogenic programs, we performed single-cell RNAsequencing of CD45+ immune cells isolated from a mouse model of SAH...

Dissociation of Murine Mammary Gland and Tumor Tissues to Create Highly Viable Single-Cell Suspensions

The dissociation of tissue into single-cell suspensions for analysis aids in understanding what is happening at the cellular level in the tumor microenvironment. A longstanding challenge in obtaining and utilizing primary cells is producing high-quality cell suspensions that preserve cell characteristics for downstream...
