TL;DR

Compared with earlier ApoC-III–targeted antisense oligonucleotides, plozasiran offers the advantages of less frequent dosing, sustained pharmacodynamic effects, and potentially a more favorable tolerability profile based on indirect evidence, positioning it as a promising therapeutic option for patients with FCS and other severe hypertriglyceridemia disorders inadequately managed with existing therapies.

Single-dose ION775, a GalNAc-conjugated siRNA targeting APOC3, lowers triglycerides for up to 12 months in moderate hypertriglyceridemia.

BACKGROUND Apolipoprotein C-III (apoC-III) regulates triglyceride-rich lipoprotein metabolism. ION775 is a GalNAc-conjugated small interfering RNA designed to silence APOC3 mRNA and enable infrequent dosing. OBJECTIVE To evaluate the safety, tolerability, and lipid effects of single-dose ION775 in adults with moderat...

Evaluating enlicitide decanoate: first oral PCSK9 inhibitor for the treatment of adults with hypercholesterolemia.

Enlicitide decanoate is a promising addition to lipid-lowering therapy, potentially expanding access to PCSK9 inhibition through oral administration, however, its definitive clinical role will depend on cardiovascular outcomes data, long-term safety, adherence in real-world practice, and cost-effectiveness compared wit...

Therapeutic plasma exchange in hypertriglyceridemia-induced acute pancreatitis: A narrative review of indications, timing, and clinical outcomes

Hypertriglyceridemia (HTG) is the third leading cause of acute pancreatitis (AP) and is associated with considerable morbidity and mortality. Rapid triglyceride (TG) reduction is a central component of treatment for hypertriglyceridemia-induced acute pancreatitis (HTG-AP). Therapeutic plasma exchange (TPE) has been inc...
