Abstract

The determination of small-molecule compounds (SMCs), including veterinary drugs, pesticides, and mycotoxins, in food and environmental matrices remains a critical challenge in analytical chemistry, often demanding detection methods with high sensitivity and reliability. Conventional immunoassays for SMCs are predominantly limited to competitive formats due to the low molecular weight and single-epitope nature of these compounds, which often results in modest sensitivity and linearity. Anti-immune complex (anti-IC) recognition elements specifically bind to neo-epitopes that are generated upon formation of the antigen-antibody complex and are absent from either component alone. This binding property overcomes the limitation that small molecules cannot be detected in a sandwich format, enabling noncompetitive immunoassays in food matrices and related samples with markedly enhanced analytical performance. This review systematically consolidates the principal strategies for generating anti-IC recognition elements, critically evaluates their integration across diverse detection platforms, and synthesizes current mechanistic insights into how these reagents recognize immune complexes. It serves as a comprehensive guide for their rational development and application in high-performance immunoassays for SMC monitoring.

Development of a Potent QS-21-Derived Adjuvant Photoaffinity Probe: Identification of Cathepsin D as the Molecular Receptor.

The saponin natural product QS-21 is a potent immunological adjuvant used in human vaccines to enhance the immune response but suffers from scarcity, instability, and dose-limiting toxicity, precluding its advancement as a stand-alone adjuvant. Further, putative cellular receptor(s) and its molecular mechanisms of acti...

Rational hapten design enables monoclonal antibody-based immunoassays for class-selective detection of phthalate esters in edible oils.

Phthalate esters (PAEs) are frequently detected in edible oils, necessitating rapid detection methods. However, the small molecular size and high structural similarity of PAEs make it challenging to develop antibodies that combine analytical sensitivity with group-selective recognition. Herein, a monoclonal antibody (m...

A review on lateral flow immunoassay technology for mycotoxin detection: From single-signal enhancement to multi-signal synergy.

Mycotoxins are toxic secondary metabolites produced by fungi that frequently contaminate grains, nuts, and other foodstuffs. Their widespread occurrence causes significant economic losses and poses a serious threat to human and animal health. Traditional analytical methods relying on complex instruments often fail to m...
