The findings reinforce the high unmet need in patients with HR+/HER2’s metastatic breast cancer whose disease has become resistant to ET, highlighting the need for effective treatments in this setting.

Abstract

To describe patient characteristics, treatment patterns and real-world survival in patients with hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2–) metastatic breast cancer (mBC) who received chemotherapy in the post-endocrine therapy (ET) setting where cytotoxic agents are the mainstay of treatment. This retrospective, observational cohort study utilized data from the US nationwide Flatiron Health Research Database. Women aged ≥ 18 years with HR+/HER2 − mBC who received chemotherapy post-ET from January 1, 2015, to December 31, 2024, were included. Patient characteristics, treatment patterns, real-world overall survival (rwOS), and time to next treatment or death (rwTTNTD) were described post-ET. 5041 patients were included; median age was 63 years at mBC diagnosis. Most were non-Hispanic White (63%) and treated in the community setting (82%) and had Eastern Cooperative Oncology Group performance status 0–1 (59%) at the start of first-line treatment (1L) post-ET, recurrent mBC (68%), and ≥ 2 prior lines of ET-based treatment in the mBC setting (65%). Median time from mBC diagnosis to 1L post-ET was 19 months. Most patients (97%) received chemotherapy 1L post-ET. Median (95% CI) rwOS from start of 1L post-ET was 13.5 (13.0–14.1) months. 30% of patients who initiated 1L post-ET did not survive to the next treatment line; there was a decrease in rwOS with each subsequent treatment. Similar findings were observed for rwTTNTD. These findings reinforce the high unmet need in patients with HR+/HER2 − mBC whose disease has become resistant to ET, highlighting the need for effective treatments in this setting.

Treatment Outcomes of Patients with Early-Stage Breast Cancer Meeting NATALEE Eligibility Criteria. A Real-World Cohort Study

The findings provide real-world historical outcome data for patients who would now be considered candidates for adjuvant ribociclib and may serve as a benchmark for evaluating outcomes following adoption of adjuvant ribociclib.

Real-world response rates in patients with human epidermal growth factor receptor 2-positive breast cancer after neoadjuvant treatment using dual human epidermal growth factor receptor 2 blockade

Introduction Breast cancer (BC) is the most commonly diagnosed malignancy among women globally and in India. The human epidermal growth factor receptor 2 (HER2)-positive subtype is associated with aggressive disease and poorer prognosis; however, neoadjuvant therapy combining dual HER2 blockade with chemotherapy has be...

Chemotherapy Use, Healthcare Resource Utilisation, and Direct Costs in High-Risk Early-Stage ER+/HER2– Breast Cancer in Italy: A Real-World Study Linking Administrative and Pathology Data

Background/Objectives: Patients with high-risk early-stage oestrogen receptor-positive/human epidermal growth factor receptor 2-negative early breast cancer (ER+/HER2– eBC) remain at high risk of recurrence despite standard endocrine therapy. Given the limited real-world data in Italy, this analysis evaluated treatment...

Trends in primary endocrine therapy utilization among patients with early-stage ER+/HER2− breast cancer who do not undergo surgery: A National Cancer Database analysis.

44 Background: Primary endocrine therapy (pET) is a mainstay of cancer treatment for patients with estrogen receptor-positive (ER+) breast cancer who are unfit for surgery. As the multidisciplinary breast oncology teams continue to personalize cancer therapies for patients, we have noticed pET being expanded to...

Predictive and Prognostic Value of Androgen Receptor Expression Across All Molecular Subtypes in Patients with Breast Cancer Who Received Neoadjuvant-Treatment: Single-Center Retrospective Analysis

Purpose Androgen receptor (AR) expression is detectable in the majority of breast cancers across all molecular subtypes; however, its predictive value for pathological complete response (pCR) and its prognostic contribution to long-term survival in an all-comer neoadjuvant chemotherapy (NACT) cohort remain incompletely...

Real-World Treatment Outcomes and Prognostic Factors in Metastatic EGFR-Mutated Non-Small Cell Lung Cancer: A Single-Center Study from a High-Volume Tertiary Chest Hospital

Background/Objectives: Epidermal growth factor receptor (EGFR)-mutated non-small cell lung cancer (NSCLC) is a distinct molecular subgroup responsive to EGFR tyrosine kinase inhibitors (TKIs). This study evaluated real-world treatment outcomes and prognostic factors in patients with EGFR-mutated metastatic NSCLC. Metho...
