Abstract

Poly(butylene adipate-co-terephthalate) (PBAT) is a biodegradable copolyester increasingly used in agricultural mulch films, yet genomic resources for PBAT-degrading bacteria remain limited. This study reports the complete genome sequence of Cupriavidus sp. PB-02, a novel strain with PBAT-degrading potential isolated from greenhouse soil in the Republic of Korea. The genome of Cupriavidus sp. PB-02 was sequenced using Oxford Nanopore long-read technology. De novo assembly yielded three circular replicons totaling 8,747,246 bp at 70.0x coverage. Average nucleotide identity analysis against 15 type strains showed a maximum of 88.86% to Cupriavidus necator, and digital DNA-DNA hybridization yielded 37.3%, supporting its classification as a novel species candidate. Structural annotation predicted 8,021 coding sequences. Functional profiling revealed 20 PBAT degradation candidate genes, 91 heavy-metal resistance genes, no antibiotic resistance genes, and 21 biosynthetic gene clusters. A total of 169 insertion sequences were detected, with 99 concentrated on the megaplasmid.
