Glia-Informed Design of Smart Biomimetic Biomaterials for Brain Repair and Neural Tissue Engineering

Sep 2026 · European Cells & Materials · 0 citations

Abstract

Smart biomimetic biomaterials are increasingly being developed to improve brain repair by providing structural support, localized therapeutic delivery, and biologically active microenvironments that promote tissue integration. However, their performance in the central nervous system is strongly influenced by the neuroimmune milieu, particularly by microglia- and astrocyte-mediated regulation of inflammation, extracellular matrix remodeling, and tissue homeostasis. This narrative review examines how glial biology can inform the design of neural biomaterials across major platform types, including hydrogels, nanoparticles, three-dimensional scaffolds, and neural interfaces. We synthesize current evidence showing that glial responses are not merely secondary reactions to implanted materials, but key determinants of biomaterial integration, regenerative permissiveness, and long-term functional compatibility. We further discuss how stimulus-responsive and extracellular matrix-mimetic materials can be engineered to sense local injury-associated signals and deliver temporally and spatially controlled therapeutic effects. Based on these observations, we outline a glia-informed design perspective centered on microenvironmental sensing, temporal regulation of neuroinflammation, biomimetic matrix architecture, and selective modulation of glial functions relevant to tissue repair. The review highlights translational considerations and biological readouts for evaluating biomaterial performance at cellular, tissue, and functional levels. By integrating advances in biomaterials science with glial biology, this article provides a biologically grounded design perspective for next-generation neural biomaterials with improved relevance for brain repair and regenerative neuroengineering.
