TL;DR

Fluorescence observation analysis indicated that the genetic stability of cloned genes was markedly improved by the Bacmid variant-derived baculoviruses with sustained expression for up to 20 passages, appropriately 2–3 times longer than the wild-type control.

CRISPR-Cpf1-Mediated T4 Phage Genome Editing for One-Step In Vivo Display of Heterologous Protein.

This engineering method allows for endogenous expression of the Soc-mCherry recombinant protein within Escherichia coli cells during phage replication, after which the recombinant protein spontaneously assembles onto the capsid of the engineered phage.

Construction of a versatile cloning vector for Agrobacterium tumefaciens-mediated transformation of Beauveria bassiana using benomyl for selection.

Beauveria bassiana is a filamentous fungus that has considerable potential to combat insect pests. Therefore, an easy to use genetic transformation tool helps to investigate which fungal genes play a role in virulence and host specificity. However, B. bassiana is often very resistant to antibiotics used as selective ag...

Modified Plasmids and Inverted Terminal Repeats Enhance Adeno-Associated Virus Production and Performance

A 140% increase in pHelper plasmid production is reported, while the most successful ITR variant increased pTransgene plasmid yields by 57% and transgene expression by 28%, without reducing rAAV yields or transduction efficiency.

Expanding Reverse Genetics of Positive-Strand RNA Viruses: Optimised Rescue Platforms and Construction of a Novel Fluorescent Reporter Nidovirus

It is demonstrated that a potent constitutive promoter system and a hammerhead ribozyme enhance the efficiency of positive-sense RNA virus rescue using CPER, and may support applications requiring efficient recovery of low fitness mutants and diverse mutational libraries and barcoded virus populations.

Effect of Improved pET Plasmids on Protein Expression Yield and Basal Expression

High-yield recombinant protein expression is a critical requirement in biopharmaceutical development. The pET vector family is among the most widely used systems for protein production in Escherichia coli. Shilling et al. (2020) reported that insertion of a GAGA sequence within the T7lac promoter region of pET28a enhan...
