Findings highlight isobavachalcone (IBC) as a potential drug candidate that promotes PD-L1 degradation and demonstrate the therapeutic potential of targeting the OTUB1-PD-L1 axis to improve immune checkpoint therapy.

Abstract

PD-L1-mediated immunosuppression is a key mechanism underlying tumor immune evasion, whereas PD-1/PD-L1 blockade therapies remain limited by low response rates and acquired resistance. Through small-molecule screening, we identify isobavachalcone (IBC), a naturally derived compound that modulates PD-L1 homeostasis by targeting the deubiquitinase OTUB1. Mechanistically, IBC targets OTUB1 at the His217 residue and suppresses its deubiquitinase activity, thereby promoting K48-linked polyubiquitination of PD-L1 and its proteasome-dependent degradation. Functionally, IBC reduces tumor cell PD-L1 expression and enhances CD8+ T cell-mediated cytotoxicity. In PD-L1-deficient tumor models, IBC fails to further enhance antitumor activity, indicating that PD-L1 is required for IBC-mediated immune regulation. Moreover, IBC combined with anti-CTLA-4 therapy further enhanced antitumor efficacy. Together, these findings highlight IBC as a potential drug candidate that promotes PD-L1 degradation and demonstrate the therapeutic potential of targeting the OTUB1-PD-L1 axis to improve immune checkpoint therapy.

Targeting ZDHHC15-mediated S-acylation of CD155 reactivates anti-tumor immunity.

The immune checkpoint CD155 is frequently upregulated across multiple malignancies and drives tumor progression and immune evasion, yet the posttranslational mechanisms regulating its stability and activity remain unclear. Here, we identify S-acylation as a key regulator of CD155 stability and immunosuppressive functio...

Targeting the PRMT5-CBX8 axis overcomes dual barriers of CD8⁺ T cell exclusion and tumor stemness for sensitizing TNBC to anti-PD-1 therapy

A novel epigenetic pathway orchestrated by PRMT5 that converges on the functional modulation of CBX8 to synchronously governs CD8⁺ T cell exclusion and tumor stemness is unveiled, nominating PRMT5 inhibition as a compelling therapeutic strategy for combination immunotherapy in TNBC.

Targeting the nonenzymatic scaffold function of RIPK1 via a VHL-recruiting PROTAC synergizes with immunotherapy in melanoma.

Receptor-interacting protein kinase 1 (RIPK1) mediates cell survival, inflammation, and cell death, and its kinase-independent scaffolding function drives tumor progression and therapeutic resistance, which cannot be eliminated by conventional RIPK1 kinase inhibitors. To overcome this limitation, we adopted proteolysis...

Targeting the PD-L1-induced PDK4/GLS metabolic axis overcomes anti-PD-1 resistance in non-small cell lung cancer

This study identifies a novel ‘PD-L1–ER stress–pyruvate–macrophage senescence’ axis as a key mechanism underlying primary resistance to ICB and proposes that targeting the PDK4/GLS-dependent pyruvate surge offers a promising therapeutic strategy to sensitize tumors to anti-PD-1 immunotherapy.

Lactate-associated H3K18la upregulates RNF166 to promote immune evasion in hepatocellular carcinoma by remodeling c-Jun ubiquitination

Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality, and PD-L1-mediated immune evasion is a major mechanism of tumor immune suppression and an important target of immune checkpoint blockade. Here, we identified RNF166 as an E3 ubiquitin ligase that promotes immune evasion in HCC. RNF166 expres...
