Abstract

The gamma delta T-cell receptor (γδ TCR) mediates γδ T-cell responses that are crucial for immune defense, tissue surveillance, and homeostasis. While recent studies suggest that γδ TCRs engage ligands multivalently, how multivalent engagement activates the γδ TCR remains elusive. Here, we present the structures of the Vγ9Vδ2 (clone G115) TCR in both the inactive and the agonist-crosslinked active states. In the γδ TCR lipoprotein complex, cholesterol and phospholipids form a “barrier” that isolates CD3ζζ from the other subunits and likely prevents autoactivation. As an agonist, the Fab fragment of the UCHT1 antibody dimerizes upon binding the Vγ9Vδ2 TCR, thereby crosslinking and activating the receptor. Cryo-EM analysis of Fab-crosslinked Vγ9Vδ2 TCR identifies three Fab dimer interfaces, one between the two light chains and the other two between light and heavy chains. These Fab-Fab interfaces, together with the Fab-CD3ε interface, mediate the assembly of the Vγ9Vδ2 TCR and UCHT1 Fab into 2:2 tetramers. Upon activation, the Vγ9Vδ2 TCR becomes conformationally plastic and loses enriched lipids, including cholesterol and phosphatidylethanolamine. Our findings reveal a dimerization-driven activation mechanism, inform the design of rigid TCR-crosslinking agonists, and suggest multivalency-induced clustering as a general paradigm for γδ TCR activation.

Butyrophilin 3A1 engages the CDR2δ loop of the Vγ9Vδ2+ T cell receptor.

It is shown that intracellular pAg accumulation can modulate binding of Vγ9Vδ2+ TCR in a BTN3A1-dependent manner, indicating the important role of TCR binding to BTN3A1 in pAg recognition.

Mechanistic basis for the selective recognition of the Fcγ receptor IIa by monoclonal antibody IV.3.

The monoclonal antibody IV.3 selectively binds the platelet Fcγ receptor IIa (FcγRIIa), potently blocking immune complex engagement without cross-reacting with the closely-related FcγRIIb. This specificity has made IV.3 invaluable for dissecting FcγRIIa-mediated activation in diverse conditions, including infection, au...

γδ T cells: bridging innate and adaptive immunity in development, tissue residency, and disease

γδ T cells are a unique lymphocyte lineage that bridges innate and adaptive immunity through MHC-unrestricted recognition of phosphoantigens, lipids, and stress-induced proteins. Their tissue-resident subsets, shaped by Vγ/Vδ chain usage, play essential roles in barrier surveillance, tissue homeostasis, and host defens...

Spatiotemporal co-regulation of IL2 and TCR receptors reveals early antigen-driven synaptic synergy in human CD8+ T cells

It is shown that TCR and IL2 signaling overlap in space and time and synergize when simultaneously triggered, demonstrating synergistic TCR and IL2R signaling that drives efficient expansion of antigen-specific CD8 + T cells.
