Findings reveal distinct physiological roles for chloroplast-localized βCAs in tomato and identify SlβCA4 as an important contributor to plant growth, photosynthesis, and temperature-dependent stomatal developmental plasticity.

Abstract

β-Carbonic anhydrases (βCAs) catalyze the reversible interconversion of CO2 and bicarbonate and contribute to plant carbon metabolism and environmental responses. However, the physiological functions of individual βCA isoforms in crops remain poorly understood. Here, we investigated two chloroplast-localized βCAs, SlβCA1 and SlβCA4, in tomato (Solanum lycopersicum). We combined subcellular localization, CRISPR/Cas9 mutagenesis, grafting, leaf gas-exchange measurements, and controlled environmental treatments to examine their roles in growth, photosynthesis, and stomatal developmental plasticity. Both proteins localized to chloroplasts but their disruption resulted in markedly different phenotypes. While SlβCA1 mutants showed largely normal growth and photosynthesis, SlβCA4 mutants exhibited severe root and shoot growth defects. Grafting mutant shoots onto wild-type rootstocks partially restored vegetative growth, indicating that impaired root function contributes to the growth defects. SlβCA4 mutants also showed reduced CO2 assimilation and stomatal conductance, while electron transport remained largely unaffected. Elevated CO2 partially improved leaflet growth but did not restore plant height or root elongation. Furthermore, unlike wild-type plants, SlβCA4 mutants failed to increase stomatal index in response to elevated temperature. These findings reveal distinct physiological roles for chloroplast-localized βCAs in tomato and identify SlβCA4 as an important contributor to plant growth, photosynthesis, and temperature-dependent stomatal developmental plasticity.

β-Carbonic anhydrases integrate CO2 and light cues to coordinate photosynthesis in tomato.

This work identifies two tomato β-carbonic anhydrases responsive to deficient CO2 and establishes chloroplast βCA1 as a key integrator of CO2 and light cues that optimizes photosystem performance through modulation of pmf partitioning and reveals stromal pH regulation as a previously unrecognized mechanism for optimizi...

α-Carbonic Anhydrase 1 Affects Photosynthetic CO2 Assimilation and Stomatal Conductance in Arabidopsis thaliana Plants

Arabidopsis thaliana plants of two mutant lines with a knockout of the At3g52720 gene, encoding stromal α-carbonic anhydrase 1, αCA1 (αCA1-KO), and wild-type plants of the Columbia ecotype (Col), were grown under 450 ppm (NCO2) and 150 ppm (LCO2) CO2 in the air. The CO2 assimilation rate (ACO2) in plants grown under NC...

Knockout of mitochondrial dicarboxylate carriers AtDIC1 and AtDIC3 affects germination, biomass accumulation, and carbon partitioning in Arabidopsis thaliana

AtDIC1 and AtDIC3 make distinct, partially overlapping contributions to mitochondrial metabolism, influencing germination performance, carbon allocation, and redox balance in Arabidopsis thaliana. The mitochondrial dicarboxylate carrier (DIC) is essential for the exchange of dicarboxylic acids across the inner mitochon...

Exogenous biotin modulates non-stomatal photosynthetic limitation under salinity in in vitro–grown Dizygostemon riparius

This study investigates whether exogenous biotin (vitamin B7) mitigates salinity-induced physiological limitations in Dizygostemon riparius, a medicinal species endemic to Maranhão, Brazil, grown in vitro. Salinity (50 mM NaCl) significantly restricts shoot growth, reduces shoot dry mass, and decreases leaf number. Gas...

Reducing transitory starch in cassava impairs photosynthesis and carbon export to storage roots

Optimizing source–sink relations is a central strategy for improving crop yield. In potato, reducing transitory leaf starch synthesis can redirect carbon to storage organs without impairing growth. To test whether this strategy is transferable to cassava, we suppressed the major small subunit of ADP-GLUCOSE PYROPHOSPHO...

A β-Mannanase Gene, MAN2, Coordinates Tomato Fruit and Seed Development and Salt Stress Tolerance.

Tomato (Solanum lycopersicum) β-mannanase is known as playing an important role in mannan degradation; however, its biological functions and transcriptional regulation remain poorly understood. Here, we characterized the roles of mannan-1,4-β-mannanase 2 (SlMAN2) in tomato development and salt stress tolerance. Results...
