These findings identify a candidate AKR1C3-associated epithelial–stromal–myeloid spatial configuration in gastric cancer, and proof-of-concept evidence of close spatial proximity among the corresponding marker-defined populations is provided.
Abstract
The spatial organization of epithelial states relative to stromal and myeloid cells in gastric cancer remains incompletely understood.
We integrated single-cell RNA-sequencing data from 36 normal and tumor tissue samples (GSE183904) with Visium spatial transcriptomics (GSE251950). Cell-state signatures were evaluated in TCGA-STAD (n = 375), and multiplex immunohistochemistry was performed on specimens from three patients.
Epi4_AKR1C3 and FB3_ACTA2 showed nominal enrichment in tumor tissues, whereas Mac2_SPP1 showed no significant compositional difference between normal and tumor tissues. Higher signature scores for these three states were nominally associated with shorter overall survival (log-rank P = 0.044, 0.019, and 0.048, respectively), although none remained significant after multiple-testing correction. Spatial deconvolution identified regions of co-enrichment, and multiplex immunohistochemistry provided proof-of-concept evidence of close spatial proximity among the corresponding marker-defined populations.
These findings identify a candidate AKR1C3-associated epithelial–stromal–myeloid spatial configuration in gastric cancer. Further mechanistic studies are needed to validate its biological and clinical significance.
INTRODUCTION
The spatial heterogeneity of the tumor microenvironment in gastric cancer, particularly in the stromal compartment, remains poorly characterized. Fibroblast activation protein (FAP)-positive cancer-associated fibroblasts (CAFs) are known to promote tumor progression, immune evasion, and therapeutic resista...

Intratumoral epithelial heterogeneity contributes to lung adenocarcinoma (LUAD) progression and may limit the interpretability of prognostic models derived from bulk tissue. To characterize this heterogeneity and assess its clinical relevance, four public single-cell RNA-sequencing datasets comprising 193,280 quality-c...

The role of growth arrest-specific 2-like 3 (GAS2L3) in cholangiocarcinoma (CCA) cells remains unclear. This study explored GAS2L3 expression, its prognostic value, and its impact on CCA cell migration. We integrated public transcriptomic data with immunohistochemistry from tumor tissues of 55 patients who underwent re...

Oxidative stress response (OSR) is suggested as a therapeutic target in triple negative breast cancer (TNBC). However, the cell type specificity and spatial distribution of OSR genes and proteins remain poorly defined. We re-analyzed public datasets, demostrating high OSR gene expression in TNBC tumor cells. We gener...
Pancreatic ductal adenocarcinoma (PDAC) is characterized by a dense desmoplastic stroma and profound spatial heterogeneity, yet the multicellular organizational principles governing its tumor microenvironment remain poorly understood. Here, we performed single-cell RNA sequencing (scRNA-seq; n = 9) and spatial transcri...

Background/Objectives: Tumor-associated macrophage heterogeneity may influence colorectal cancer (CRC) outcomes. We investigated a CTSD-associated macrophage program and its functional relevance. Methods: Single-cell GSE132465 data (23 tumor and 10 normal specimens), TCGA colorectal expression and survival data, and th...
