Biology

TL;DR

A cryopreservable and scalable workflow is developed to generate hiPSC-derived hepatic organoids that can be directly matured to either static HLOs or LoC systems, enabling matched comparison of both platforms and establishing a robust hiPSC-derived LoC platform.

A human iPSC-derived kidney–liver organ-on-a-chip platform for modeling inter-organ crosstalk

The liver and kidneys are essential for drug metabolism and clearance and are highly sensitive to toxicity. Most in vitro models assess these organs in isolation, limiting the study of inter-organ interactions. Here, an induced pluripotent stem cell (iPSC)-derived kidney–liver organ-on-a-chip model was developed that c...

Reverse bioengineering of the liver: developmental principles for next-generation liver-on-a-chip systems

Despite major advances in liver-on-a-chip and organoid technologies, most current in vitro liver models remain limited. Here, it is argued that these limitations are fundamentally conceptual rather than purely technical. Reverse bioengineering is introduced as a unifying design framework for liver-on-a-chip systems, in...

Mesenchymal stem cell-derived liver-on-a-chip reconstitutes human liver structure and function.

An in vitro model of human liver with functional integrity would advance liver research and enable personalized therapeutics. A microfluidic liver-on-a-chip can generate a functional hepatobiliary network using human liver cells and recapitulate blood circulation and bile excretion. However, many current microfluidic l...

Behaviour of iPSC-Derived and Donor-Derived Hepatic Stellate Cells in Organoid Cultures Is Highly Variable and Contributes to Fibrotic Outcome

Hepatic stellate cells (HSCs) are central drivers of liver fibrosis, yet their heterogeneity is often overlooked in human liver organoid models. Here, we investigated how HSC-intrinsic variability influences fibrotic phenotypes in multicellular liver organoids using induced pluripotent stem cell-derived HSCs (iHSCs) an...

Human Liver Organoid Platforms for MASH Drug Discovery: Evaluating Immunometabolic Fidelity, Pharmacological Predictivity, and Translational Readiness

Metabolic dysfunction-associated steatohepatitis (MASH) is a progressive form of metabolic dysfunction-associated steatotic liver disease characterised by steatosis, inflammation, hepatocellular injury and fibrosis. Human liver organoid platforms offer a controllable system for modelling selected MASH-related processes...

Integrated analyses of in vivo mouse models and liver organoids reveal MFG-E8-mediated protection against hepatocyte injury.

Milk Fat Globule-EGF Factor 8 (MFG-E8) is a multifunctional glycoprotein implicated in tissue homeostasis and repair. While its roles in intestinal and pancreatic injury are well established, its function in hepatic regeneration remains poorly defined. In this study, we investigated MFG-E8 expression and activity using...
