TL;DR

Results showed that L. helveticus R0052 reduced liver injury and inflammation, and ameliorated cholestasis in DDC-fed mice, and provided an experimental basis for the potential application of L. helveticus R0052 in the prevention of cholestasis.

Abstract

Cholestasis is an important clinical manifestation of primary sclerosing cholangitis (PSC), leading to liver injury, fibrosis and liver failure. The gut microbiota influences the pathogenesis of cholestatic liver disease by modulating metabolic and immune responses. Our study established a PSC mouse model with a 0.1% 3,5-diethoxycarbonyl-1,4-dihydrocollidine (DDC) diet and Lactobacillus helveticus R0052 intervention to assess the protective effects of probiotics on the disease. In addition, we investigated the microbiome and transcriptome alterations resulting from L. helveticus R0052 intervention to explore associations between gut microbiota and host metabolism. The results showed that L. helveticus R0052 reduced liver injury and inflammation, and ameliorated cholestasis in DDC-fed mice. These beneficial effects were accompanied by alterations in hepatic FXR-Cyp7a1 and ileal FXR-Fgf15 signaling pathways, modified bile-acid homeostasis, as well as changes in the gut microbiota and their metabolites. Overall, our research findings provide an experimental basis for the potential application of L. helveticus R0052 in the prevention of cholestasis.

Development and clinical validation of Lactobacillus helveticus BGRA43: An innovative probiotic approach for MASLD

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common chronic liver disease worldwide and is closely associated with metabolic disorders such as obesity, insulin resistance, and dyslipidemia. Its prevalence continues to increase, and current management strategies rely primarily on lifestyl...

Tetramethylpyrazine ameliorates metabolic dysfunction-associated steatohepatitis by modulating gut microbiota dysbiosis and restoring intestinal barrier integrity.

BACKGROUND AND PURPOSE This study investigated the therapeutic effects and gut-liver axis-related mechanisms of tetramethylpyrazine (TMP) in metabolic dysfunction-associated steatohepatitis (MASH). EXPERIMENTAL APPROACH Male C57BL/6 mice were fed a methionine- and choline-deficient (MCD) diet for 6 weeks to establish...

Phocaeicola dorei YGMCC0564 attenuates HFD-induced glycolipid metabolic disorders in association with remodeling of the gut microbiota and bile acid profiles

Background Glycolipid Metabolic Disorders (GLMD) are characterized by imbalances in glucose and lipid homeostasis, often presenting with severe clinical symptoms. Current pharmacotherapies such as orlistat and metformin show clear efficacy but are associated with long-term tolerability concerns. This study aimed to eva...

Live combined bifidobacterium attenuates rivaroxaban-associated colonic bleeding in mice: evidence for a substantial gut microbiota-dependent component

Rivaroxaban, a direct oral anticoagulant (DOAC), is widely prescribed for the prevention and treatment of thromboembolic disorders. However, its use is associated with an increased risk of gastrointestinal bleeding, the mechanisms of which remain incompletely understood. Effective strategies to relief such bleeding are...

Dietary flavonoids target the gut-liver axis for the management of cholestatic liver injury.

Cholestatic liver injury (CLI) is a progressive liver disorder characterized by impaired bile acid (BA) synthesis, transport, and excretion. This dysfunction leads to the intrahepatic accumulation of toxic BAs, which in turn triggers hepatic oxidative stress, inflammation, and subsequent fibrogenesis. Through enterohep...

Total glucosides of paeony in cirrhosis: mechanisms of action via the gut–liver axis – a review

Liver cirrhosis, the end-stage of chronic liver diseases, imposes a high global burden. Gut–liver axis dysregulation—microbiota imbalance, barrier failure, endotoxin translocation, bile acid disturbance, suppressed FXR signaling, and NLRP3 activation—drives cirrhosis progression. Total glucosides of paeony (TGP), a...
