EZH2 mutation status predicts inferior outcomes after bendamustine‐based immunochemotherapy in frontline follicular lymphoma: A GELTAMO multicenter validation study
Abstract
Follicular lymphoma (FL) is the most common indolent lymphoma, 1,2 a biologically and clinically heterogeneous entity with a diverse genetic landscape and characterized by slowly progressive but relapsing clinical course. 3 The t(14;18) translocation driving BCL ‐ 2 overexpression is present in 85% – 90% of patients 4 although additional genetic events are required for lymphomagenesis. Next ‐ generation sequencing (NGS) studies have identified recurrent mutations in chromatin ‐ modifying genes ( KMT2D , CREBBP , and EZH2 ). 5,6 EZH2 encodes the catalytic subunit of the Polycomb repressor complex 2 and mediates H3K27 methylation.
