TL;DR

WT1-TCR-T cells co-expressing CD8-41BB receptors were able to completely regress established cell line-derived ovarian tumors, showed robust in vivo expansion and persistence, and provided long-term protection from tumor rechallenge, suggesting this platform can be a universal approach for enhancing the therapeutic potential of TCR-based cell therapies.

CAR T cell-induced interferon gamma enhances MHC class I expression and sensitizes neuroblastoma to TCR-engineered T cell therapy

T cell receptor (TCR)-engineered T cells target antigens presented on major histocompatibility complex (MHC) class I molecules. MHC class I expression is often low in childhood tumors such as neuroblastoma but can be stimulated by interferon gamma (IFNγ). Chimeric antigen receptor (CAR) T cell effector function, in...

Engineering T cells in vivo to express a synthetic cytokine receptor enables preferential expansion and enrichment of anti-CD22 CAR T cells.

Preclinical studies are described demonstrating that UB-VV400 generates anti-CD22 CAR T cells that kill CD22-expressing B cells and tumor cells in vitro and in vivo, and that engagement of RACR results in the selective expansion and enrichment of CAR T cells in vivo leading to complete tumor clearance and B cell deplet...

Nanoscale artificial antigen presenting cells to expand human antigen-specific cytotoxic CD4 T cells for immunotherapy 2258982

CD4 T cells play key roles in immune modulation and cancer immunity. Increasing evidence shows they also exhibit cytotoxicity and can be harnessed for cancer immunotherapy. However, methods to reliably expand antigen-specific CD4 T cells remain limited. Here, we developed a nanoscale platform to expand antigen-sp...

Tumor-specific CD4 T cells cooperate with myeloid cells to remodel the pancreatic tumor microenvironment and enable effective immunotherapy

We interrogate antigen-specific CD4 T cells during immunotherapy in pancreatic ductal adenocarcinoma. Vaccination with MHC-II-restricted tumor epitopes impart superior protection compared to an immunodominant MHC-I epitope, prompting development of MHC-II affinity-enhanced tetramers to track tumor-specific CD4 T cells....

Activity in vivo of in vitro expanded and adoptively transferred antigen-specific CD8+ T cells against neoepitopes that mediate tumor rejection 2306826

A subset of somatic passenger mutations in cancer cells lead to generation of peptides which can be presented by MHC I molecules and recognized by CD8 T cells. Such peptides are tumor- specific and are termed neoepitopes. A small subset of neoepitopes can immunize mice against tumors in models of prophylaxis and...

MHC-II-restricted neoantigen expression drives tumor infiltration, activation, and maintenance of nonspecific T cells in lung cancer 2251169

Cancer mutations can result in the expression of tumor-specific “neoantigens” that are targeted by T cells. Neoantigen expression drives robust T cell infiltration of tumors across cancer models, yet most of these T cells are not specific for the neoantigens and contain both pathogenic and immunosuppressive subse...
