Abstract

Although flavan-3-ols are extensively utilized as acrylamide inhibitors in thermal food processing, their variable efficacies suggest a more complex, unresolved mechanism. Overcoming the traditional perspective of simple inhibition, this study reveals a paradoxical, structure-dependent dual effect of four typical flavan-3-ols (EGCG, EGC, ECG, and EC) on acrylamide formation. While the net inhibitory efficacy in an asparagine-glucose system followed EGCG > EGC ≈ ECG > EC, investigations using a 3-aminopropionamide (3-APA) intermediate model demonstrated that the oxidation of the flavan-3-ol B-ring generated highly reactive quinone intermediates that actively facilitated acrylamide formation. This promotional capacity (EC > ECG > EGC > EGCG) inversely mirrored the net inhibitory trend. Crucially, UHPLC-MS/MS analysis tentatively identified nine novel quinone-amine adducts, providing structural evidence for this B-ring-mediated promotion. Consequently, the net impact of flavan-3-ols on acrylamide formation is not a simple linear inhibition, but a dynamic competition between A-ring-mediated carbonyl trapping and B-ring-mediated quinone generation. EGCG exhibits the most potent net inhibition because its highly substituted structure is exceptionally susceptible to competing side reactions, such as autoxidation and ester hydrolysis, which effectively quench the B-ring-driven promotion. These findings provide direct mechanistic and practical guidance for selecting galloylated tea polyphenols to mitigate acrylamide risk in thermally processed starchy foods.

Phase-dependent antioxidant behavior in a synthetic C-α-glucogallin analogue library by photoredox-mediated α-ketonylation.

Phenolic acid glycosyl esters like glucogallins are promising antioxidants but are often limited by hydrolytically labile O-glycosidic linkages. Here, twelve C-α-glucogallin analogues were synthesized in six steps from commercial peracylated sugars by combining four monosaccharides with three plant-derived phenolic aci...

Sphingofungin-based aminopolyols and DIX-like azasugars - their synthesis and in vitro biological profile.

A simple stereodivergent approach to sphingofungin-based aminopolyols and their modification into 1,5-dideoxy-1,5-iminoxylitol (DIX)-based 1-N-iminosugars was accomplished. The use of a [3,3]-heterosigmatropic rearrangement on carbohydrate scaffolds, followed by Grubbs metathesis chemistry, provided an efficient route...

Non-chelating 2-(arylthio)acetophenone derivatives as broad-spectrum tyrosinase inhibitors: Synthesis, in silico profiling, in vitro, and in vivo antimelanogenic efficacy.

A series of 2-(arylthio)acetophenone (2-ATAP) derivatives (1-12) were designed and synthesized as novel tyrosinase inhibitors (TYRIs). Among them, five derivatives exhibited superior mushroom tyrosinase (mTYR) inhibitory activities compared to kojic acid, with derivative 5 demonstrating a potent nanomolar IC50 value. D...

Synthesis, Z/E Isomerism, and Cytotoxic Activity of β-Hydroxy Oximes Derived from Indenoquinoxalinone, Tryptanthrin, and a Tricyclic Isatin Analogue: A Combined Experimental and DFT Study

Indolo[2,3-b]quinoxalines and their N-substituted derivatives exhibit anticancer activity. In this work, a convenient two-step protocol comprising base-promoted aldol condensation followed by oximation was used for the synthesis of novel β-hydroxy oximes from heterocyclic ketones. The oximes were obtained as Z/E mixtur...

N-(3'-(Dimethylamino)propyl)amides based on 18βH-glycyrrhetinic acid retain P-glycoprotein inhibitory and MDR-reversing activities.

Multidrug resistance (MDR) is a major obstacle to successful cancer chemotherapy, primarily mediated by P-glycoprotein (P-gp/ABCB1), which actively effluxes structurally unrelated anticancer drugs. We previously reported Sol-DMAP, an 18βH-glycyrrhetinic acid-derived P-gp inhibitor that reverses chemoresistance and exhi...

Mechanistic exploration of 4-bromobenzyloxy-1,2,3-triazole derivatives as dual cholinesterase inhibitors: Kinetic, molecular dynamics, and halogen-contact insights.

Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) regulate cholinergic neurotransmission, and their partial inhibition is an established symptomatic strategy in Alzheimer's disease; clarifying how individual ligands engage these enzymes is therefore central to inhibitor design. To probe these cholinesterase-...
