Findings indicate that the TAG structure influences digestive kinetics and acute whole-body metabolic responses, supporting the further evaluation of MLCT for nutritional applications involving age-related digestive decline.
Abstract
Age-related declines in gastrointestinal function increase the malnutrition risk and highlight the need for lipid formulations with digestive and metabolic characteristics suited to older populations. In this study, a structurally characterized medium- and long-chain triacylglycerol (MLCT) prepared by enzymatic transesterification was compared with a compositionally equivalent physical blend (PB) using a simulated in vitro gastrointestinal digestion model for the elderly and an in vivo indirect calorimetry in adult mice. MLCT exhibited a more gradual fatty acid release pattern and 33-44% lower lipolysis during the initial gastrointestinal stages than PB without affecting final hydrolysis, and it significantly improved polyunsaturated fatty acid bioaccessibility. In mice, MLCT administration was associated with comparatively gradual changes in whole-body energy expenditure and respiratory gas exchange among the lipid treatments. These findings indicate that the TAG structure influences digestive kinetics and acute whole-body metabolic responses, supporting the further evaluation of MLCT for nutritional applications involving age-related digestive decline.
Omega-3 polyunsaturated fatty acids (n-3 PUFA), such as α-linolenic acid (ALA), are crucial for human health. Although n-3 PUFAs in food are mainly in triacylglycerol (TAG) form, they are often provided as ethyl esters (EE) in supplements. Previous research has demonstrated improved postprandial bioavailability of n-3...

Although lipid-based formulations (LBFs) of self-emulsifying drug delivery systems (SEDDS) have been used for poorly water-soluble drugs, they have many limitations, the first being the low solubility of drugs in lipids, which limits drug loading in formulations, and the second being inadequate dispersion of formulatio...

Abstract: Dietary proteins and lipids coexist in foods in diverse structural forms, and their interactions influence lipid digestion and the subsequent intestinal absorption of lipolysis products, including free fatty acids and monoacylglycerols. This review examines the role of protein–lipid interactions (PLIs) in lip...

Pectin is indigestible by human enzymes and is metabolized in the human gastrointestinal tract (GIT) by gut microorganisms to yield short-chain fatty acids (SCFAs) and other metabolites. Pectin ingestion has been shown to lower blood cholesterol levels and low-density lipoprotein cholesterol, without changing levels of...
