Circulating lymphocytes in HNSCC display coordinated expression of selected senescence-associated genes, with IL-6 and CXCL1 as candidate prognostic biomarkers linked to tumor progression that warrant further validation.

Abstract

Senescence-associated secretory phenotype (SASP) signaling, along with key markers such as P16INK4a/CDKN2A and LMNB1, has not been systematically studied in circulating lymphocyte subsets in head and neck squamous cell carcinoma (HNSCC). This study aimed to evaluate SASP-related genes and senescence markers in peripheral CD4+, CD8+, and CD19+ cells and assess their clinical relevance. Expression of IL-6, IL-1β, TNFα, CXCL1, P16INK4a/CDKN2A, and LMNB1 was measured by RT-qPCR in sorted lymphocytes from 58 HNSCC patients at baseline, 31 post-treatment, and 13 controls. Statistical analyses included nonparametric tests, correlation analyses, and survival models (Kaplan–Meier, Cox regression). In the results, LMNB1 was significantly upregulated in all lymphocyte subsets of HNSCC patients. IL-6, CXCL1, and IL-1β were elevated in CD4+ T cells. A coordinated co-expression network involving IL-6, CXCL1, IL-1β, P16INK4a/CDKN2A, and LMNB1 was observed. Clinically, IL-6 in CD8+ T cells was associated with higher nodal stage and worse survival, while CXCL1 in CD19+ B cells independently predicted survival. No differences were found between pre- and post-treatment samples. Circulating lymphocytes in HNSCC display coordinated expression of selected senescence-associated genes, with IL-6 and CXCL1 as candidate prognostic biomarkers linked to tumor progression that warrant further validation.

Prognostic Relevance and Immune Correlates of DAPK1 Expression and CD4+/CD8+ T-Cell Infiltration in Oral Squamous Cell Carcinoma

It is suggested that DAPK1 is associated with apoptosis-related signaling, increased immune-cell infiltration, and favorable clinical outcomes in OSCC, although its independent prognostic value requires validation in larger cohorts.

Prognostic impact of HLA-G expression in cervical squamous cell carcinoma: correlation with PD-L1 and CD8-positive tumor infiltrating lymphocytes

Background Purpose of the study was to investigate the prognostic impact of HLA-G expression in patients with cervical squamous cell carcinoma (CSCC), and to further analyze its correlations with PD-L1 and CD8-positive (CD8+) tumor-infiltrating lymphocytes (TILs) within the tumor microenvironment (TME). Methods A total...

Increased density of CD103-positive tissue-resident memory T cells predicts favorable response to immunotherapy in oral cancers.

Immune checkpoint inhibitors (ICIs) have become an important therapeutic option for oral cancers; however, reliable biomarkers predicting treatment efficacy remain limited. In this study, we investigated the association between intratumoral immune cell subsets, regional lymph node (RLN) immune status, and clinical resp...

Tumor CD19 Expression Determines Distinct Clinical Outcomes After CD19 CAR T-Cell Therapy in Large B-Cell Lymphoma.

It remains uncertain whether lower CD19 expression is clinically relevant for outcomes of CD19 CAR-T therapy of large B-cell lymphoma (LBCL). We conducted an integrative analysis of tumor CD19 levels with centralized quantitative assessment by flow cytometry (FC), immunohistochemistry and RNA-sequencing in a LBCL cohor...

Combined PD-L1/CD8 immune phenotyping in breast cancer: exploratory associations across molecular subtypes and insights into possible exhaustion-related immune dysfunction

Background Programmed death-ligand 1 (PD-L1) expression and CD8+ tumor-infiltrating lymphocyte (TIL) density are established biomarkers of tumor–immune interaction in breast cancer; however, the biological and prognostic significance of their combined assessment across molecular subtypes remains incompletely defined. T...
