The protein kinase IKK epsilon shapes the melanoma tumor microenvironment by modulating angiogenesis and the T-cell immunity

Inhibitor of nuclear factor kappa-B kinase epsilon (IKKε) contributes to tumorigenesis and metastasis in various cancers. In melanoma, it is overexpressed and constitutively active. Inhibition of IKKε by either knockdown or pharmaceutical compounds reduce tumor progression by suppressing key molecular pathways within m...

USP22 is a novel vulnerability regulating MEIS1 protein abundance and gene transcription in KMT2Ar acute leukemia.

A genome-wide CRISPR/Cas9 screen in a KMT2Ar B-cell acute lymphoblastic leukemia cell line identified the deubiquitinase USP22 as a novel regulator of MEIS1 protein stability, that could potentially be exploited as a therapeutic target in the future in KMT2Ar leukemias.

Phosphorylation Protects Oncogenic RAS from LZTR1-Mediated Degradation.

PAK1 and PAK2 shielded RAS from LZTR1-dependent degradation by phosphorylating T148, and inhibiting PAK1/2 activity improved RAS-directed therapy, revealing a regulatory circuit governing RAS stability that is preferentially active in blood cancers and potentially druggable.
