Functional evaluation of TCR-pMHC pairs at scale allows in silico TCR reactivity prediction.

Accurate prediction of T cell receptor (TCR) reactivity is a long-standing goal in immunology. Here, we asked whether functional validation of TCR-peptide-major histocompatibility complex (MHC) (TCR-pMHC) pairs at scale alters performance estimates of TCR-pMHC reactivity prediction models. We developed a TCR rapid asse...

A genome-wide CRISPR screen in human prostate cancer cells reveals drivers of macrophage-mediated cell killing and positions AR as a tumor-intrinsic immunomodulator

Immunity protection from macrophages is revealed as an adverse consequence of hormonal therapy in prostate cancer patients using a genome-wide co-culture CRISPR screen and positions tumor-intrinsic AR signaling as a bona fide immunomodulatory pathway.

Molecular Glue and Phosphorylation-Dependent 14-3-3 Recruitment to p53 with an Engineered Interaction Cassette.

These results establish a modular, MG- and phosphorylation-dependent platform for engaging 14-3-3 in a p53 context and for evaluating how chemical stabilization translates from biochemical interaction control to cellular pathway-level readouts.
