Abstract
Transketolase (TKL; EC 2.2.1.1) has been validated as a potential new herbicide target. To develop a new and highly effective herbicides-targeted TKL, a series of indole-containing fusarium acid (FA) derivatives 9a-9s were designed and synthesized. Among them, compound 9o exhibited more excellent herbicidal activity against Digitaria sanguinalis and Amaranthus retroflexus (inhibition > 90%) compared to the positive controls at 375 g ai/ha. Furthermore, compound 9o was safe for maize and wheat (phytotoxicity 5%) at 375 g ai/ha. Also, compound 9o exhibited a slightly better inhibition effect against Setaria viridis (Sv)TKL than that of the positive control (IC50: 0.229, 0.420 mg/L). Molecular docking and molecular dynamics simulations (MDS) indicated that the binding between SvTKL and 9o is stronger. Moreover, SvTKL-9o has more favorable binding residues including SER101, ARG169, PHE512, HIS539, and ARG598. These results have guiding significance for the molecular design of higher herbicidal activity compounds targeting TKL.
Acetohydroxyacid synthase (AHAS, EC 2.2.1.6) is a core enzymatic target in agrochemical research for herbicide development and has been widely investigated in recent decades. To develop novel AHAS-targeted herbicides, twenty-three acylthiourea derivatives were synthesized via fragment recombination and bioisosteric rep...

The application of herbicides plays a critical role in ensuring food security. However, the long-term overuse of herbicides can lead to a range of adverse effects. Therefore, it is essential to develop innovative and sustainable herbicides. A series of pyridazinone derivatives targeting phytoene desaturase (PDS) were d...

Enhancing innovation in herbicide discovery is imperative to combat weed resistance and safeguard global food security. To develop novel herbicide targeting dihydroorotate dehydrogenase (DHODH), a series of 3-carboxy-4-pyrazole amide lead scaffold bearing ester or acid substituents were designed and synthesized. Bioa...

The use of herbicides represents one of the effective weed management strategies. To create high-efficiency, low-risk protoporphyrinogen IX oxidase (PPO)-targeted herbicides, a set of novel N-phenylphthalimides bearing 1,3,4-oxadiazole and acylhydrazide fragments was designed and synthesized. Bioassays revealed that mo...

Protoporphyrinogen oxidase (PPO) is one of the major targets for herbicide discovery. N-Phenylpyrazole derivatives bearing ester, oxime, and isoxazoline moieties were synthesized via isomerization, derivatization and cyclization. Bioassays showed C34 potently inhibited Nicotiana tabacum PPO (NtPPO) (Ki = 15.6 nM), wi...
