TL;DR
A29, safe, potent multi-mechanistic lead, exerts multimodal anti-Xoo effects by disrupting key processes, virulence, and promising as novel agrochemical for BLB.
Abstract
Background
Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), severely threatens rice production and global food security. Current commercial bactericides such as thiadiazole copper (TC) and bismerthiazol (BT) are losing efficacy due to widespread Xoo resistance, while research on structural modification and activity optimization of indole derivatives for Xoo control remains scarce, highlighting an urgent need for novel, safe, and effective anti-Xoo agents and filling this research gap.
Results
Twenty-nine indole derivatives (A1-A29) were synthesized from 3-carboxyindole (structures confirmed by 1H NMR, 13C NMR, HRMS). A29 showed superior anti-Xoo activity (EC50 = 1.24 μg/mL), low phytotoxicity, high selectivity, and better greenhouse efficacy (51% curative, 50% preventive) than BT and TC. Mechanistically, A29 exerts its effects through multiple pathways that include EPS and biofilm modulation, ROS-mediated apoptosis induction, and regulation of key genes, as validated by RT-qPCR.
BACKGROUND
Plant diseases pose a persistent threat to agricultural and forestry production, while the extensive use of conventional fungicides has promoted resistance development and raised environmental residue concerns. Therefore, developing plant essential oil-derived antifungal leads with novel modes of action repr...

BACKGROUND
Plant bacterial diseases caused by Xanthomonas oryzae pv. oryzicola (Xoc) and Xanthomonas axonopodis pv. citri (Xac) cause severe yield losses, and increasing resistance to existing bactericides demands novel antibacterial agents. Guided by a molecular hybridization and cationization strategy, a series of py...

BACKGROUND
The persistent proliferation of bacterial phytopathogens presents a significant threat to global food security and agricultural sustainability. Among these pathogens, species belonging to the genus Xanthomonas are particularly destructive. Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of bacterial le...

Plant pathogenic fungi pose a significant threat to global crop productivity and food security. Herein, we report the design and synthesis of 55 coumarin–oxazole, −isoxazole, and −pyrazole hybrids as novel antifungal agents. Most derivatives exhibited notable potency, with C1, C4, and C5 showing strong activity again...

A total of twenty pyrazole derivatives were synthesized using alkoxy or halo acetophenones in reaction with N, N-dimethylformamide dimethylacetal (DMF-DMA). These compounds were characterized through 1H NMR, 13C NMR, and LC-HRMS techniques. Their herbicidal efficacy against barnyard grass (Echinochloa crus-galli) was e...
