Abstract
The main aim of this investigation was to improve the
technique of developing doubled haploids in Tagetes
erecta L. by anther culture and thereby doubling the
chromosomes. Heat shock pre-treatment was given to
the anthers of both marigold genotypes 'Desi Orange'
and 'Desi Yellow' post culturing on enriched MS (EMS)
medium. The efficient and early development of initial
callus and shoot buds were observed when anthers
were induced on EMS medium supplemented with
meta-topolin (0.5 mg/l) and NAA (0.25 mg/l). Ploidy
levels were determined by counting the number of
chromosomes and chloroplasts in stomatal guard cells.
The addition of oryzalin at a concentration of 5 µM
exposed for 48 hours proved to be more effective in
achieving the maximum frequency of chromosomal
doubling. This refined protocol can be applied to
marigold to expedite the production of doubled
haploids.
Polyploidy induction is widely used in ornamental breeding to generate novel phenotypes such as larger flowers, thicker stems, and increased vegetative biomass. Therefore, this study aimed to evaluate oryzalin-mediated chromosome doubling in in vitro Culture of Dahlia pinnata using a completely randomized 2×6 factorial...

In transgenic orchids, the effect of a transgene on flower color cannot be scored until the plant flowers, which may take three to five years after transformation. Inducing flowers in vitro would allow for much earlier screening. Therefore, we tested whether plant growth regulators (PGRs) added to a modified Knudson C...

Ruscus hypoglossum L. is an evergreen ornamental species valued as cut foliage, but its commercial production is constrained by slow vegetative propagation and limited availability of uniform planting material. This study aimed to develop an efficient in vitro propagation protocol for R. hypoglossum by evaluating the e...
