This conceptual review examines the highly interconnected, bidirectional crosstalk between the molecular circadian clock, glial-mediated neuroimmunity, and the macroscopic glymphatic clearance system and suggests that neurodegeneration can also be viewed as neurodegeneration as a network-wide collapse of biological timing in addition to a localized cellular failure.
The glymphatic system is a brain-wide fluid and solute transport pathway that contributes to cerebrospinal fluid-interstitial fluid exchange and the removal of metabolites, inflammatory mediators, and aggregation-prone proteins. Glymphatic activity is shaped by sleep state, vascular pulsatility, respiration, vasomoti...

The glymphatic system has been proposed as a brain-wide perivascular clearance pathway involved in the exchange of cerebrospinal fluid and interstitial fluid. As a dynamic clearance network, it plays key roles in regulating fluid exchange between the cerebrospinal and interstitial compartments, facilitating the removal...

INTRODUCTION
Maintaining homeostasis in the Central Nervous System (CNS) requires constant, bidirectional molecular interactions between microglia and astrocytes. This intercellular communication, mediated by cytokines, extracellular vesicles, purinergic signals, and connexin hemichannels, synchronizes immune surveilla...

This review critically evaluates the experimental, epidemiological, and mechanistic evidence linking early-life metabolic and environmental insults with the establishment of latent vulnerability and proposes that this status may remain clinically silent for decades until activated by aging and cumulative stressors, ult...

Preclinical data support the notion that modulation of AQPs activity may offer neuroprotection through restoration of homeostasis and reduction of inflammation in neurodegenerative disease, and its potential and limitations as a therapeutic target for the prevention of CNS disorders are described.

This review examines the molecular and cellular mechanisms through which neuroinflammation contributes to the progression of AD, with particular emphasis on microglial activation, TREM2 and soluble TREM2 (sTREM2) signaling, NLRP3 inflammasome activation, astrocyte–microglia interactions, and blood–brain barrier dysfunc...
