TL;DR

While both LprG-Rv1410c and MmpL11 pathways are involved in modulating cellular c-di-GMP levels during biofilm formation, only LprG-Rv1410c are important for regulating GPL synthesis and/or surface localization, and MmpL11 may play a broad role in fine-tuning GPL levels.

Functional characterization of GplA (MSMEG-0394) reveals a novel determinant of glycopeptidolipids homeostasis in Mycobacterium smegmatis

Introduction Glycopeptidolipids (GPLs) are major constituents of the cell envelope of non-tuberculous mycobacteria (NTM) and play essential roles in colony morphology, surface motility, biofilm formation, antimicrobial susceptibility, and host–pathogen interactions. Although the GPL biosynthetic locus has been extensiv...

PPE57 Cooperates with MmpL3 to Mediate Bacterial Lipid Transport and Promote Persistent Infection of Mycobacterium tuberculosis

These findings establish PPE57 as an essential accessory component of the MmpL3 lipid transport system, bridging mycobacterial cell wall assembly with host nutrient exploitation during persistent infection.

Functional dissection of the smi1 regulator delineates its pivotal roles in modulating cellular physiology, Monascus pigments production and citrinin accumulation in Monascus purpureus.

Results indicate that smi1 not only participates in cell wall biosynthesis and morphological regulation in M. purpureus, but also potentially regulates the synthesis of MPs and citrinin by affecting central and secondary metabolic networks.

Mycobacteriophage Adephagia gp48 inhibits essential host cell wall biosynthesis enzyme mannose-1-phosphate guanylyltransferase

It is shown that gp48 encoded by mycobacteriophage Adephagia is toxic when overexpressed in Mycobacterium smegmatis and Mycobacterium abscessus and the toxicity results from gp48 interaction with the host mannose-1-phosphate guanylyltransferase (Mpg) enzyme.

Recognition and mechanism of M. tuberculosis cell wall galactan transport

Mycobacterial-derived lipid-linked galactan (LLG) is an essential component of the Mycobacterial tuberculosis (Mtb) cell wall core arabinogalactan (AG), a well-established virulence factor and target of anti-tubercular drugs. Cross-membrane translocation of LLG is energetically unfavorable and is therefore catalyzed by...
