Results indicate that ZmSk2 is a key component of mitochondrial RNA splicing complexes and contributes to mitochondrial homeostasis and maize kernel maturation.

Abstract

Splicing of group II introns in the organelles of higher plants requires nuclear-encoded protein co-factors. However, the exact mechanisms and interactions among these auxiliary proteins are not yet established. In the present work, we characterized a maize kernel mutant, zmsk2. The growth of endosperm and embryo in the zmsk2 mutant was severely arrested during kernel development. ZmSk2 is a mitochondria targeted CRM-domain protein that functions in splicing of group II introns, particularly nad2 introns 2 and 4 and nad5 introns 2 and 4. There were no mature nad2 and nad5 transcripts in zmsk2 which was associated with aberrant assembly of mitochondrial complexes I and I + III and then with considerable decrease in the activity of NADH dehydrogenase. Protein-protein interaction experiments revealed that ZmSk2 formed homodimers and interact with Zm-mCSF1, Dek37 and PPR231. Taken together, these results indicate that ZmSk2 is a key component of mitochondrial RNA splicing complexes and contributes to mitochondrial homeostasis and maize kernel maturation.

The splicing factor SPR2 interacts with the RNA-binding protein PPR8 to facilitate mitochondrial intron splicing and is essential for maize seed development.

Intron splicing plays a crucial role in organellar gene expression in plants, yet its mechanism remains poorly understood. We previously reported a small PPR protein, SPR2, which is a general splicing factor in the splicing of over half of the mitochondrial introns via interaction with PPR-SMR1, a small MutS-related do...

The maize CMS-C fertility restore gene ZmRf4b encodes a SPL transcription factor that represses ZmMAP1D-mediated co-translational turnover of mitochondrial toxic ATP6C protein.

The promising CMS-C/Rf system has not been applied on a large scale in maize hybrid breeding due to the unclear restoration mechanism for the primary restorer ZmRf4b. In CMS-C maize, the mitochondrial chimeric gene atp6c encodes a cytotoxic variant of ATP6 that over-accumulates and disrupts the assembly of mitochondria...

The conserved U1 snRNP subunit FgLuc7 regulates sexual development and pre-mRNA processing in fusarium graminearum.

The U1 small nuclear ribonucleoprotein (U1 snRNP) is an essential spliceosome component that mediates pre-mRNA splicing. Although the molecular function of Luc7 proteins is broadly conserved in eukaryotes, whether this core splicing factor acquires developmentally specialized functions in filamentous fungi remains uncl...
