Tyrosol notably enhanced sperm performance, suggesting its potential integration into clinical strategies for AZS management and fertility preservation and the dysregulation of the metabolite tyrosol, which is associated with AZS.

Abstract

Male factor infertility accounts for approximately 50% of all infertility cases. Metabolomics has recently been applied to the assessment of sperm quality and male fertility. However, studies on the biological functions and clinical effects of sperm metabolites in relation to asthenozoospermia (AZS) remain limited. In this study, we analyzed the metabolomic profiles of seminal plasma from 20 men with AZS and compared them with those from 20 men with normal semen parameters (healthy controls) using a standardized untargeted high-performance liquid chromatography-tandem mass spectrometry approach. Metabolite levels differed significantly between the AZS and healthy control groups. A total of 177 potential biomarkers and multiple metabolic pathways were identified in men with AZS. Specifically, tyrosol levels were significantly reduced in the AZS group. Preincubation with tyrosol significantly increased the percentage of spermatozoa with progressive motility in men with AZS. Moreover, tyrosol supplementation improved progressive motility, DNA integrity, and acrosomal integrity in cryopreserved sperm samples from these individuals, as reflected by post-thaw sperm parameters. In conclusion, this study reveals the distinctive metabolic characteristics of seminal plasma and the dysregulation of the metabolite tyrosol, which is associated with AZS. Tyrosol notably enhanced sperm performance, suggesting its potential integration into clinical strategies for AZS management and fertility preservation.

Alterations in urinary microbiota composition and function in men with Asthenozoospermia: a case-control study

Asthenozoospermia (AZS) is a leading cause of male-factor infertility, yet the contribution of the urinary microbiota to sperm motility remains poorly defined, because previous work has focused almost exclusively on the seminal and gut microbiota. Here we used 16S rRNA gene sequencing to characterize the composition an...

Exploratory seminal plasma metabolomics and independent HPLC–UV evaluation in men with impaired semen quality

Background Impaired semen quality may involve concurrent abnormalities in sperm concentration, total sperm number, motility, morphology, and DNA integrity. The corresponding alterations in the seminal plasma metabolome remain incompletely characterized. Objectives To characterize seminal plasma metabolic alterations in...

Sodium valproate at concentrations bracketing estimated seminal plasma levels impairs human sperm function in vitro through oxidative stress and mitochondrial dysfunction.

Sodium valproate (VPA), a first-line broad-spectrum antiseizure medication, has been associated with altered semen quality in clinical settings, but its direct effects on human sperm remain incompletely defined. In this in vitro study, sperm from normozoospermic donors were exposed to VPA concentrations bracketing an e...

Untargeted Metabolomic Profiling of Spermatozoa and Seminal Plasma Identifies Candidate Metabolic Indicators of Semen Quality in Holstein Bulls

This exploratory study applied untargeted LC–MS/MS metabolomic profiling to investigate metabolic differences between Holstein bulls with high-quality (HQ) and low-quality (LQ) semen in both spermatozoa and seminal plasma. Bulls were classified based on long-term routine semen quality records and ejaculate rejection ra...
