Ten Years of Ocrelizumab in Relapsing Multiple Sclerosis
Abstract
Key Points Question What is the long-term safety and efficacy profile of ocrelizumab in patients with relapsing multiple sclerosis (RMS)? Findings In the OPERA I/II randomized clinical trials including 1656 patients with RMS, treatment with ocrelizumab for up to 10 years led to near-complete and sustained suppression of relapse activity, robust control of progression independent of relapses, and minimal disability accrual. Patients starting ocrelizumab 2 years earlier had significantly greater relapse and disability control compared with patients initially treated with interferon beta-1a, without new or cumulative safety signals emerging over the 10-year follow-up. Meaning Results confirm the long-term clinical benefits and manageable safety profile of ocrelizumab and reinforce the importance of early initiation of high-efficacy treatment in patients with RMS.
