Abstract

EL-RAKHAWI CLOSED-LOOP AUTONOMOUS PLATFORM V2.0: PRECISION THERAPEUTICS & VACCINE DISCOVERY Author: Dr. Mohamed Kamal Arafa El-RakhawiDOI: 10.5281/zenodo.23346881 A revolutionary closed-loop autonomous platform solving the 90% clinical failure rate in drug discovery by integrating generative AI, self-driving labs, and organ-on-a-chip microphysiological systems, bridging the translational gap between in silico design and human-relevant validation. Core architecture: Generative foundation model (with synthetic accessibility pre-filtering) + self-driving robotic synthesis lab + microphysiological evaluation array (Body-on-a-Chip) + reinforcement learning orchestrator with surrogate temporal neural network + multi-fidelity active learning funnel. Four critical operational corrections: (1) Multi-Fidelity Funnel implemented - 1000 candidates generated, filtered by synthetic accessibility, top 20 synthesized and biochemically screened, only top 3 evaluated in Organ-on-a-Chip, solving throughput bottleneck; (2) Surrogate Temporal Network processes high-dimensional time-series MPS data (calcium transients, cytokine kinetics) into dense reward vectors, replacing oversimplified scalar endpoints; (3) Synthetic accessibility ($S_{synth}$) applied as hard constraint in generative model before synthesis, preventing resource waste on infeasible compounds; (4) Claim 6 added protecting multi-fidelity funnel mechanism, preventing competitor workarounds. Performance: Reduces discovery timeline from 3-5 years to 6-12 months, eliminates animal testing dependency, enables rapid vaccine design within days of pathogen sequencing. Applications: accelerated drug discovery ($20B), rapid vaccine design ($15B), personalized medicine ($10B). Total market: $45B+ by 2030. Comprehensive IP protection: 6 technical claims covering closed-loop integration, surrogate temporal network reward processing, multi-fidelity funnel, synthetic accessibility pre-filtering, Body-on-a-Chip configuration, and autonomous discovery method.
