Biology

TL;DR

It is proposed that an inactivating conformational change is induced when dynein detaches against strain from its catch-bonded state, and captured the dynamics of this process through a statistical model of Entropically Constrained Reactivation, obtaining predictions in excellent agreement with experimental data.

Energetics of the Spontaneous Beating of Motor-Driven Filaments

Cilia and flagella beat through the collective action of molecular motors, driving fluid transport and cell motility, and are an attractive template for engineered actuators. The follower force model captures spontaneous beating in minimal reconstituted systems, but treats motor activity as a uniform, constant force de...

KobA mediates kinesin-1-dependent transport of dynein-dynactin to microtubule plus ends

Cytoplasmic dynein transports diverse cargoes toward microtubule minus ends following activation by the dynactin complex and cargo adapters. In elongated cells such as fungal hyphae and neurons, dynein must first undergo kinesin-1-dependent transport to microtubule plus ends to engage cargoes, but the mechanism linking...

The Molecular Interplay of Tau and Motor Proteins on Microtubules

Tau is a microtubule (MT)-associated protein essential for neuronal cytoskeletal organization and intracellular transport, yet how tau physically regulates motor proteins remains elusive. Here, we integrate single-molecule imaging, cryo-electron microscopy, and molecular dynamics (MD) simulations to investigate the mol...

Microtubule organisation by motors and nonmotor microtubule-associated proteins.

Microtubules are key components of the eukaryotic cytoskeleton. These stiff polymers often span the entire cell, provide structural support, and act as tracks for long-distance transport and force generators. Their organisation underlies the logistics of intracellular cargo transport, resistance to compressive loads, a...

Spatial relief of Kinesin-1 auto-inhibition by Ensconsin regulates ncMTOCs localization and shapes microtubule networks.

During Drosophila oogenesis, the organization of microtubules (MTs) drives polarized transport and cytoplasmic flows. Although Kinesin-1 plays a central role in these processes, its molecular and spatial regulation remains poorly understood. Here, we show that Ensconsin, an activator of the Kinesin heavy chain (Khc), i...
