Circular RNAs (circRNAs) are an emerging class of RNA molecules involved in development and disease. They are generated through back-splicing from pre-mRNA. RNaseR treatment, which degrades linear RNA while preserving circRNAs, is widely used for circRNA validation and enrichment. This short report demonstrated differential performance of two RNaseR enzymes from different suppliers, despite identical experimental conditions, indicating that these enzymes should not be considered interchangeable across suppliers. Importantly, these findings highlight that without demonstrating efficient linear RNA degradation, unchanged circRNA abundance after RNase R treatment is inherently uninformative and may contribute to false-positive circRNA validation or inadequate circRNA enrichment. We therefore highlight that RNaseR digestion should be consistently verified using a linear control, particularly when changing enzyme suppliers
This review explores the current understanding of intrinsic molecular features dictating circRNA stability and provides an integrated perspective for understanding circRNA-centered regulation of RNA stability.

Two complementary methods for tsRNA validation are presented, small RNA northern blotting, which allows direct visualization of RNA size and abundance, and dumbbell PCR, which enables isoform-specific quantification with single-nucleotide resolution, which provide robust tools to validate tsRNA expression and facilitat...

Circular RNAs (circRNAs) are generated by backsplicing of eukaryotic protein-coding transcripts and can regulate microRNAs and RNA binding proteins, or serve as translation templates. Their covalently closed structure confers resistance to exonuclease-mediated degradation, extending their half-life and supporting their...

Circular RNAs (circRNAs) are covalently closed RNA molecules generated through back-splicing of precursor mRNA (pre-mRNA) transcripts. Although circRNAs lack the 5' cap and the 3' poly(A) tail characteristic of linear mRNAs, their circular structure confers enhanced stability and resistance to exonucleolytic degradatio...
