TL;DR

Following inhaled and oral dosing, londamocitinib demonstrated rapid absorption, and faecal excretion as the principal route of elimination and exhibited an acceptable safety and tolerability profile.

Abstract

Aim

Londamocitinib (AZD4604) is an inhaled, selective Janus kinase 1 (JAK1) inhibitor in development for asthma. This open-label, two-part, phase I clinical study (NCT06157255) used a radiolabelled microtracer approach to evaluate the absolute bioavailability and the absorption, distribution, metabolism and excretion (ADME) of inhaled londamocitinib after intravenous and oral administration in healthy male and female participants (n = 8).

Methods

Pharmacokinetics, safety, metabolic profile and metabolite identification from plasma, urine, and faecal samples were assessed employing highly sensitive accelerator mass spectrometry. Participants received a single oral 4-mg dose of [14C]londamocitinib (NMT 37.0 kBq [14C]) in Part 1 and an intravenous 30-μg dose of [14C]londamocitinib (NMT 37.0 kBq [14C]), followed by a single inhaled dose of 3-mg londamocitinib in Part 2.

Results

Mean recovery of total radioactive material after oral dosing was 76.3% in faeces and 8.7% in urine. After intravenous dosing, recovery was 67.3% in faeces and 16.0% in urine. Absolute bioavailability of oral londamocitinib was 40.4% and for inhaled administration was 62.5%. Unchanged londamocitinib constituted approximately 70% of circulating radioactivity in plasma and accounted for 47.8% and 28.4% of the administered dose in excreta after oral and intravenous administration respectively. No new safety signals were observed.

Conclusion

Following inhaled and oral dosing, londamocitinib demonstrated rapid absorption, and faecal excretion as the principal route of elimination. Unchanged londamocitinib and metabolite M6 were the major entities present in plasma, urine and faeces. Londamocitinib, at these doses, exhibited an acceptable safety and tolerability profile.

Pharmacokinetic and bioequivalence evaluation of teneligliptin hydrobromide tablets following single-dose administration in healthy Chinese volunteers under fasting and fed conditions

By inhibiting dipeptidyl peptidase-4 (DPP-4), teneligliptin provides a therapeutic option for type 2 diabetes mellitus (T2DM) that is widely used in Asia. A generic product could lessen treatment costs, provided that its systemic availability is equivalent to that of the branded formulation. Such comparative informatio...

Pharmacokinetics and Safety of Deuremidevir Hydrobromide (JT001) in Subjects with Mild and Moderate Renal Impairment: An Open-Label, Parallel-Group Phase I Study.

JT001 (deuremidevir hydrobromide, VV116) is an oral antiviral prodrug that is rapidly converted in vivo to its active metabolite, 116-N1. Since 116-N1 is mainly cleared by the kidneys, renal impairment may increase its systemic exposure. This study evaluated the pharmacokinetics of 116-N1 following a single 0.3-g oral...

Safety, pharmacokinetics, and pharmacodynamics of intravenously administered deuterated N,N-dimethyltryptamine (CYB004) in healthy volunteers.

BACKGROUND Serotonergic psychedelics, such as N,N-dimethyltryptamine (DMT), have shown therapeutic potential in various psychiatric disorders. However, DMT demonstrates pharmacokinetic (PK) variability and a short half-life. AIMS This exploratory study investigated the PK, pharmacodynamic (PD), and safety profile of...
