Abstract
Iglesias-Herrero et al. longitudinally assess JAK-inhibitor treatment response in two patients with STAT1 gain-of-function and show that it induces rapid but incomplete immune reconstitution, with increased TCR diversity and reversal of monocyte hyperinflammation, highlighting both therapeutic benefit and persistent immune dysregulation in this inborn error of immunity.
