Abstract
Abstract The Arabidopsis thaliana PHOSPHATE 1 (PHO1) was first identified as an inorganic phosphate (Pi) exporter mediating Pi loading into the root xylem. PHO1 is the prototypical member of a family of eukaryotic proteins possessing both an SPX and EXS domain. Recent cryo-EM structures of both the Arabidopsis PHO1 homolog PHO1;H1 and its mammalian ortholog Xenotropic and Polytropic Retrovirus Receptor 1 (XPR1), revealed that these proteins work as Pi channels that use a double-gated mechanism controlled by inositol polyphosphate. Key amino acids involved in binding Pi and gating the channel are highly conserved in PHO1 homolog throughout eukaryote superphyla, including in unicellular organisms, indicating a possible conservation of function during evolution. Beyond their structural homology, both plant and animal PHO1 ortholog are primarily localized to intracellular vesicles, raising interesting questions about the cell biology of Pi export. Arabidopsis PHO1 also contributes to the response of guard cells and seeds to abscisic acid (ABA) through yet unknown mechanisms. In this review, we will highlight how the recently gained knowledge about PHO1 and XPR1 structure and its cell biology is shaping our views about how these proteins contribute to Pi homeostasis at the cellular and organismal levels.
Protein import across the chloroplast outer membrane is mediated by the TOC complex and regulated by GTP hydrolysis at two receptor GTPases, large and small. The former are the β-barrel-containing Toc159 family of proteins (Sun and Jarvis 2023; H. Li and Chiu 2010; Shi and Theg 2013a; Kessler and Schnell 2009). The sma...

Eukaryotic cells store inorganic phosphate in the form of long chains called polyphosphates (polyP) that can reach many hundreds of residues in length. In the budding yeast Saccharomyces cerevisiae, polyP is synthesized by the VTC complex using ATP as a substrate and sequestered at high concentrations in the vacuole. H...

Inositol pyrophosphates are conserved signaling molecules synthesized by the bifunctional PPIP5K enzymes, but how their cellular functions diversify across species remains poorly understood. Here, we compared the PPIP5K enzyme Asp1 in the fission yeasts Schizosaccharomyces pombe and Schizosaccharomyces japonicus and in...

It is demonstrated that PsORP1 of Phytophthora sojae specifically localises to ER–Golgi membrane contact sites (MCSs) and is indispensable for vegetative growth and asexual/sexual development of P. sojae and elucidates that oxathiapiprolin directly targets the ORD to competitively block this lipid exchange, providing a...
