TL;DR

It is demonstrated that influenza virus initiates genome selection in the nucleus and reveal nuclear export as an early step in genome selection, and a promoter-dependent checkpoint is identified that links RNA promoter architecture and polymerase conformation to RNP export and packaging.

A tripartite mechanism licenses RNA polymerase import into the phage nucleus

Nucleus-forming jumbo phages import a phage-encoded non-virion RNA polymerase (nvRNAP) into a proteinaceous nucleus, but how it is selected for import is unknown. Here, we identify an import pathway that couples nvRNAP subunit interactions with phage-encoded import factors. Maximal nvRNAP import requires a novel factor...

Influenza A Virus RNA Polymerase Is Enriched on the Chromatin of Innate Immune Response Genes

It is demonstrated how FluPol accumulation across key cellular defense loci spans from initiation through to termination, driving host transcriptional attenuation and contributing to widespread defects in transcription termination.

Structural insights into the early transcription factors of the African swine fever virus.

African swine fever virus (ASFV) relies on a virus-encoded transcription machinery to initiate gene expression within cytoplasmic viral factories. The viral early transcription factor (VETF), a heterodimer composed of pD1133L and pG1340L, is required for early transcription and viral replication, yet its structural org...

Ebola virus mRNAs contain RNA structures that are critical for viral infection and targetable by antisense oligonucleotides

The first in-cell secondary structure maps of four EBOV mRNAs are presented, finding that structure content is concentrated within protein coding regions, resembling the highly structured genomes of positive-sense RNA viruses, while untranslated regions are relatively unstructured.

Novel conserved RNA structural motifs regulate genome cyclization of yellow fever virus

The long-range RNA-RNA interaction between the termini of a flavivirus genome is controlled by multiple local RNA secondary and tertiary structures. However, a complete picture of this essential genome cyclization process is still elusive. Herein, by using selective 2′-hydroxyl acylation analyzed by primer extension (S...

IRES Mutation Confers a Replicative Advantage to SVA by Enhancing Translation Efficiency

This study aimed to characterize the mutational and evolutionary patterns of the SVA genome during its transmission within host cells exhibiting enhanced innate immunity, and indicated that the mutant virus was unable to evade host antiviral innate immunity, whereas it significantly enhanced the translational activity...
