Flexible and cost-effective nature of the diatomite/AgNPs substrate, combined with its strong SERS enhancement, offers significant advantages over traditional immunoassay techniques.
Abstract
In this study, we used flexible and inexpensive diatomite/AgNPs strips and Layer-by-Layer (LbL) assembly to develop sandwich assays for the detection of cancer biomarkers of interest via Surface-enhanced Raman scattering (SERS). The modified SERS active strips were used to capture antigens of interest, incubate Raman probes, and generate SERS maps based on specific Raman peaks of the Raman probes. Simultaneous detection and quantification of multiple biomarkers, including HER2, CA15-3, CA27-29, PSA, and MUC4, were demonstrated by the immobilization of specific antibodies and the use of uniquely labeled Raman probes. Spatially resolved and co-immobilized multiplexing strategies were developed and successfully validated, showing high specificity and sensitivity with negligible background interference from serum components. The flexible and cost-effective nature of the diatomite/AgNPs substrate, combined with its strong SERS enhancement, offers significant advantages over traditional immunoassay techniques.
Surface-enhanced Raman scattering (SERS) has been widely applied to food safety screening owing to its high sensitivity and fingerprint recognition. However, SERS faces challenges in practical applications related to the precise control of the number of hot spots and the determination of how many of them actually contr...

Surface-Enhanced Raman Spectroscopy (SERS) allows for ultrasensitive detection of analytes in complex biofluids. This capability is particularly important when measuring analytes at low concentrations or when clinical sample volumes are limited. SERS improves workflows by eliminating complex sample-preparation st...

Circular RNAs (circRNAs) have recently emerged as promising biomarkers for the early diagnosis of cancer and the development of targeted therapeutic strategies. However, their detection remains challenging due to their low abundance and the difficulty in discriminating them from their linear RNA counterparts. Herein, a...

Surface-enhanced Raman spectroscopy (SERS) enables the sensitive, label-free, and multiplexed detection of biomarkers in biofluids. Flexible SERS substrates, in particular, facilitate non-invasive, on-site biofluid measurements. This talk summarizes the application of flexible SERS substrates for the detection of bioma...
Accurate discrimination between prostate cancer (PCa) and benign prostatic hyperplasia (BPH) remains clinically challenging. Here, we developed a multiprobe serum surface-enhanced Raman scattering (SERS) sensing platform comprising Ag NPs (Ag), 4-mercaptobenzoic acid (MBA)@Ag, 4-mercaptophenylboronic acid (MPBA)@Ag, an...
